摘要:目的观察健步通络熏蒸液对兔膝骨关节炎关节软骨中细胞外信号调节激酶1/2(ERK1/2)表达的影响,探讨其关节软骨保护作用机制。方法 26只新西兰大白兔,随机抽取6只作为空白组,余20只采用改良Hulth法制作骨关节炎模型,术后4周随机抽取2只验证造模是否成功,18只随机分为模型组、熏洗组和降钙素组。模型组采用自来水熏洗,熏洗组采用健步通络熏蒸液熏洗,每次30 min,每日1次;降钙素组给予降钙素5 U/kg肌肉注射,每日1次,疗程20 d。疗程结束后行空气栓塞致死,取关节软骨观察其大体形态,PCR检测软骨中ERK1/2含量。结果与空白组比较,模型组关节面损伤严重,ERK1/2含量显著升高;熏洗组和降钙素组关节面损害较轻,且ERK1/2表达降低;熏洗组与降钙素组比较,2组关节面损害及ERK1/2表达无明显差异。结论健步通络熏蒸液对膝骨关节炎关节软骨具有保护、抑制软骨退变的作用,其机制可能与抑制ERK1/2表达有关。
Objective To investigate the effects of Jianbu Tongluo Fumigation Fluid on the expression of extracellular signal-regulated kinase1/2 in articular cartilage of rabbit knee osteoarthritis; To discuss its related mechanism of action. Methods Six New Zealand white rabbits were randomly selected among 26 as blank group and 20 were made into osteoarthritis models by modified Hulth method. After four weeks, two rabbits were chosen to verify whether modeling was successful; 18 were randomly divided into model group, fumigation group and calcitonin group. The rabbits in model group and fumigation group were fumigated with water and Jianbu Tongluo Fumigation Fluid respectively, 30 min each time, 1 time a day. The rabbits in calcitonin group were treated with calcitonin intramuscular injection of 5 U/kg, 1 time a day. The treatment lasted twenty days. After twenty days, all rabbits were sacrificed through air embolism. The general configuration of articular cartilage was observed and PCR technique was used to detect the content of ERK1/2. Results Compared with the blank group, joint surface damage in the model group was more serious and the content of ERK1/2 increased. Joint surface damage in the fumigation group and calcitonin group was relative light, and the expression of ERK1/2 decreased in articular cartilage. Calcitonin group and fumigation group had no significant difference in joint surface damage and the expression of ERK1/2. Conclusion Jianbu Tongluo Fumigation Fluid can protect and inhibit the cartilage regression of knee osteoarthritis, and the mechanisms is associated with inhibition the expression of ERK1/2.
