摘要:目的研究母婴分离应激是否可诱发母鼠抑郁样行为。方法按照体重将母鼠随机分为2组:正常对照组和模型组,每组10只。模型组小鼠在其分娩后给予母婴分离应激,母鼠与仔鼠每日分离3 h,连续14 d;正常组母鼠与仔鼠不进行母婴分离应激。以蔗糖消耗量实验和强迫游泳实验考察母婴分离应激对母鼠行为学的影响;用液质联用技术测定脑组织中五羟色胺(5-HT)含量,以免疫印迹法测定脑组织中色氨酸羟化酶的表达。结果母婴分离应激后7,14 d,模型组母鼠蔗糖消耗百分比分别为(55.8±15.7)%和(52.6±10.5)%,显著低于正常组母鼠的蔗糖消耗量(73.7±8.2)%和(73.3±11.7)%;母鼠强迫游泳不动时间:模型组明显高于正常对照组。模型组小鼠大脑的皮质、海马及额叶中5-HT含量分别为(4.68±0.95),(8.12±0.98),(6.46±0.72)ng·mg^(-1),均显著低于正常对照组;模型组小鼠大脑的皮质、海马及额叶中色氨酸羟化酶表达的灰度值分别为0.86±0.02,0.69±0.02和0.87±0.03,均显著低于正常组小鼠。模型组与正常对照组比较,以上指标差异均有统计学意义(P<0.01,P<0.001)。结论母婴分离应激可诱导母鼠出现抑郁症典型症状,并伴有脑组织中5-HT合成减少和含量减低。
Objective To investigate the effect of maternal infant sepa- ration stress on depression -like behavior in mother mice and further explore the underlying mechanism. Methods C57BL/6 mice were ran- domly divided into normal control group and model group, each group had ten mouse. Mice in model group were suffered with maternal infant separation stress (M[SS) after delivery, namely, the pups were taken away from the mother mice for 3 hours per day for consecutive 14 days. While mice in normal control group did not conduct MISS. Depression - like behavior was evaluated by sugar consumption test and forced swi- mming test. The 5 -hydroxytryptamine (5 -HT) content in the brain ti- ssue was determinate by HPLC -MS technology, and tryptophan hydroxy- lase expression in brain was detected by Western - blot. Results At 7 and 14 days after MISS, the percentage of sugar consumption in model group were (55.8± 15.7)% and (52.6 ± 10. 5)%, respectively;it sig- nificantly reduced compared to that of the normal control group (73.7± 8.2) % and ( 73.3 ± 11.7 ) % ). And the immobility time of forced swim test in model group were significantly prolonged compared to that of the normal control group, it indicating that MISS can induced depression -like behavior in mother mice. The 5 - HT content in cortex, hippocampus, and frontal lobe of model group were (4. 68 ± 0. 95 ), ( 8. 12± 0. 98 ) , (6. 46 ±0. 72)ng. mg-1, respectively; it markedly reduced when compared to that of the normal group. And the expression of tryptophan hydroxylasein cortex, hippoeampus, and frontal lobe of model group were 0. 86 ± 0. 02, 0. 69 ± 0. 02 and 0. 87± 0. 03, respectively; it also significantly lower compared to that of the normal group ; it indicating that MISS can decrease 5 - HT content and tryptophan hydroxylase expression in the brain tissue of mother mice. Compared with normal control group and model group, the above factors had significantly (P 〈 0. 01, P 〈 0. 001 ). Conclusion MISS can induce classical symptoms of depression in mother mice accompanied with decreased 5 - HT synthesis and reduced 5 - HT content in the brain tissue of stressed mice.
