摘要:目的研究四逆散干预创伤后应激及睡眠障碍的电生理机制。方法 SD大鼠50只均分为5组:空白组、生理盐水组、模型组、实验组、对照组。于应激造模前1 h,实验组灌胃四逆散水煎液(含生药0.24 g·mL^(-1)),对照组给与盐酸帕罗西汀水溶液(0.42 mg·mL^(-1))灌胃(10 m L·kg^(-1)),生理盐水组以及模型组给予等体积生理盐水,每天1次,连续7 d。用电流刺激法制备创伤后应激大鼠模型。用在体多通道神经信号采集(M-NEMEA)技术检测海马CA1/CA3区神经信号。结果神经元平均放电频率,模型组CA1/CA3(2.03/1.99 Hz)明显低于生理盐水组CA1/CA3(8.96/9.62 Hz),表明幽闭电击能明显抑制海马区神经元放电频率。实验组CA1/CA3(7.63/7.87 Hz)和对照组CA1/CA3(6.96./7.90Hz)的放电频率明显高于模型组,表明四逆散以及盐酸帕罗西汀能明显改善神经元放电活动。结论四逆散、盐酸帕罗西汀对大鼠海马区神经元放电频率有显著的正向调节作用。
Objective To study the function mechanism of the Sinisan on hippocampal neuron discharge frequency in rats with post-traumatic stress and sleep-disorderrat. Methods SD rats were divided into 5 groups,blank group,NaCl group,model group,control group,test group.Sinisan Decoction( 0. 24 g·mL^-1) and paroxetine solution(0.42mg·mL^-1),administered orally,the normal control group were given equal volume of distilled water at 1 h after established model. One times a day,continuous 7 d. Theneural coding on frequency in hippocampal CA1/CA3 were detected by the M- NEMEA after the treatment. Results The CA1/CA3( 2. 03 /1. 99 Hz) in model group is lower than CA1 /CA3( 8. 96 /9. 62 Hz) in Na Cl group. While CA1 / CA3( 7. 63 /7. 87Hz)in test group and CA1 / CA3( 6. 96. /7. 90 Hz) in control group are significantly higher than model group. It shows that Sinisan and paroxetine can improve the discharge activity of neurons. Conclusion The Sinisan and paroxetine have significant positive effect on hippocampal neuron discharge frequency.
