摘要:目的建立甘肃省肉苁蓉、沙苁蓉、盐生肉苁蓉、兰州肉苁蓉的超高效液相色谱(UPLC)指纹图谱及4种指标性成分含量测定,并进行肉苁蓉药材浸出物含量测定,为肉苁蓉药材的质量评价提供理论依据。方法按照2020年版《中国药典》一部中的醇溶性浸出物下的冷浸法测定浸出物含量;采用色谱柱ACQUITY UPLC?BEH shield RP 18(2.1 mm×100 mm,1.7μm),流动相乙腈-0.1%甲酸水溶液梯度洗脱,体积流量0.1 mL·min^(-1),检测波长330 nm,柱温35℃。结果醇溶性浸出物含量为36%~68%;松果菊苷、肉苁蓉苷A、毛蕊花糖苷、2'-乙酰基毛蕊花糖苷在2μg进样量与峰面积积分值线性关系良好(r≥0.9972),RSD≤3.82%。结论该方法操作简便、精密度高、结果可靠,可用于肉苁蓉、沙苁蓉、盐生肉苁蓉、兰州肉苁蓉的质量评价。
OBJECTIVE To establish UPLC method for the determination of echinacoside,cistanchis glycosides A,verbascoside and 2'-acetyl verbascoside in Herbra Cistanches.METHODS The content of the extract was determined by cold immersion method under the alcohol-soluble extract in the first edition of Chinese Pharmacopoeia 2020.The chromatographic column ACQUITY UPLC BEH SHIELD RP 18(2.1 mm×100 mm,1.7μm)was used.Mobile phase acetonitrile-0.1%formic acid aqueous solution gradient elution;volume flow rate of 0.1 mL·min^(-1).The detection wavelength is 330 nm.The column temperature is maitained at 35℃.RESULTS The content of alcohol soluble extract was 36%~68%.Echinacea glycosides,cistanchis glycosides A,2'-acetyl verbascoside have a good linear relationship(r≥0.9996)with RSD≤3.69%.CONCLUSION The method is simple to operate,high precision and reliable.UPLC can be used for simultaneous determination of four index components in Herbra Cistanches.
