摘要:目的:分析椎动脉型颈椎病(CSA)模型大鼠下丘脑组织差异基因表达谱变化,探讨下丘脑对CSA的调控机制.方法:将10只SPF级雄性Wistar大鼠随机分为模型组和正常组,采用复合造模法建立CSA模型,模型成功后取下丘脑组织,提取总RNA,用全基因芯片检测基因表达谱,筛选差异基因,本体论(GO)、信号通路分析.结果:与正常组比较,差异基因表达谱分析显示:差异基因总数为246个(|fold change|>2,P<0.05),其中表达上调的基因有150个,下调的有96个;富集的GO功能共有1 301条,涉及对血管形成、神经细胞功能、对外界刺激、细胞黏附、细胞增殖及细胞膜活性的调节等;参与调节的信号通路共有109条,包括脂类代谢、Ras、有丝分裂原活化蛋白激酶(MAPK)、血小板活化、环-磷酸腺苷(c AMP)、FcγR-介导的吞噬作用信号通路等.结论:下丘脑对CSA的调控机制主要是对血管形成、神经功能、对刺激的应激反应等的调节.
Aim: To analyze the differences in gene expression profile of hypothalamus tissue in CSA model rats and to explore the control mechanism of adrenal gland on CSA. Methods: Ten SPF male Wistar rats were randomly divided into model group and blank group. The CSA model was established by compound modeling method. The total RNA was extracted from the hypothalamus,and the gene expression profile was detected by whole gene chip,Gene Ontology( GO),signal pathway analysis. Results:Compared with the normal group,the difference gene expression profile analysis showed that the total number of the different genes was 246( | fold change | > 2,P < 0. 05),there were 150 up-regulated genes and 96 down-regulated genes. A total of 1301 genes are rich in GO function,related to the regulation of Angiogenesis,nerve cell function,external stimulation,cell adhesion,cell proliferation,cell membrane activity and so on; There are 109 signal transducers,including Lipid metabolism,Ras,MAPK,platelet activation,c AMP,Fc gamma R-mediated phagocytosis signal pathway and so on. Conclusion: The regulation of hypothalamus on CSA is mainly through the Vascular formation,neurological function,andstress response to stimulation and so on.
