摘要:目的:利用网络药理学方法及分子对接从整体观念探讨扶正抑瘤汤治疗肺腺癌的作用机制。方法:通过TCMSP数据库检索筛选扶正抑瘤汤中的四味中药的有效成分并其靶点,利用GeneCards数据库检索肺腺癌的疾病基因,绘制Venny图获取交集基因,利用Cytoscape 3.8.0软件获取重要蛋白,构建成分–靶点–疾病–通路网络。利用R语言进行GO富集分析和KEGG富集分析。结果:扶正抑瘤汤四味中药共筛选出17个有效成分,涉及基因194个。涉及的生物学过程主要包括对脂多糖的反应等条目;涉及的分子功能主要包括受体配体的活性等条目;涉及细胞组成主要集中在膜筏板等条目。其作用机制与血脂和动脉粥样硬化等通路有关。结论:本研究初步揭示了扶正抑瘤汤多靶点整体调节肺腺癌的作用特点,为进一步开展扶正抑瘤汤治疗肺腺癌作用机制的研究提供了新的思路和研究方法。
Objective: To explore the mechanism of the treatment of lung adenocarcinoma by using network pharmacological methods and molecular docking. Methods: The active components of the four Chi-nese herbs were selected and targeted by TCMSP database, the disease genes of lung adenocarci-noma were retrieved by GeneCards database, Venny map was drawn, and Cytoscape 3.8.0 software was used, and the component-target-disease-pathway network was constructed. The GO enrichment analysis and the KEGG enrichment analysis were performed using the R language. Results: A total of 17 active ingredients involved 194 genes. The biological processes involved include the reaction to LPS, the molecular functions including the activity of the receptor ligand, and the cell composition focusing on membrane rafts. The mechanism of action is related to pathways such as blood lipids and atherosclerosis. Conclusion: This study preliminarily revealed the role of multiple target points of zheng tumor suppression soup in regulating lung adenocarcinoma, and provided new ideas and research methods for the treatment mechanism of lung adenocarcinoma.
