摘要:目的:探讨滋阴润燥丸对干燥综合征模型小鼠颌下腺炎症及功能的作用。方法:将40只8周龄NOD/Ltj小鼠随机分为5组:模型组、西药组及滋阴润燥丸高中低剂量组,每组8只。另ICR小鼠8只为空白组。适应性喂养1周后开始干预。高、中、低剂量组分别予以1.78、0.89、0.45mg/kg的滋阴润燥丸灌胃,西药组予以60mg/kg羟氯喹灌胃,空白组及模型组均给予等量无菌蒸馏水灌胃,每日1次,连续8周。期间第0、2、4、6、8周监测小鼠饮水量。干预结束后,取小鼠颌下腺和血清。HE染色法观察颌下腺组织病理情况;ELISA法检测血清中炎性细胞因子IFN-γ、IL-10、TNF-α的水平。RT-qPCR法检测小鼠颌下腺组织中AQP5、M3R mRNA表达水平;Western blot法检测小鼠颌下腺组织中AQP5、M3R的蛋白表达;免疫组化法检测颌下腺组织中AQP5蛋白表达水平。结果:与正常组比较,模型组小鼠饮水量明显增加(P<0.05),小鼠血清中IFN-γ、TNF-α表达显著升高(P<0.05),IL-10表达显著降低(P<0.05);AQP5、M3R mRNA、蛋白表达显著降低(P<0.05);与模型组相比,西药组、滋阴润燥丸高中剂量组小鼠饮水量明显减少(P<0.05),血清中IFN-γ、TNF-α表达显著降低(P<0.05),IL-10表达显著升高(P<0.05),AQP5、M3R mRNA、蛋白表达显著升高(P<0.05)。结论:滋阴润燥丸能够降低干燥综合征模型小鼠颌下腺炎症反应,改善颌下腺分泌功能。
AIM:To investigate the effect of ZiYin RunZao(ZYRZ)pill on the inflammation and function of submandibular gland in mice modeled with Sj?gren's syndrome.METHODS:Forty 8-week-old NOD/Ltj mice were randomly divided into five groups:model group,western medicine group,and the high,middle and low dosage groups of ZYRZ,with eight mice in each group.Another 8 ICR mice were blank group.The intervention was started after 1 week of acclimatization feeding.In the high,medium and low dose groups,1.78,0.89 and 0.45 mg/kg of ZYRZ pills were administered by gavage,respectively;in the western medicine group,60 mg/kg of hydroxychloroquine was administered;and in the blank group and the model group,equal amounts of sterile distilled water were administered by gavage once a day for eight consecutive weeks.The body weight and water intake of the mice were monitored at weeks 0,2,4,6 and 8.At the end of the intervention,mice submandibular gland and serum were taken,and the histopathology of submandibular gland was observed by HE staining;the levels of inflammatory cytokines IFN-γ,IL-10,and TNF-αwere detected in serum by ELISA;the levels of AQP5 and M3R mRNA were detected in submandibular gland by RT-qPCR;the levels of AQP5 and M3R mRNA in submandibular gland were detected in submandibular gland by Western blot;and the levels of AQP5 and M3R mRNA in submandibular gland were detected in submandibular gland by RT-qPCR.AQP5,M3R protein expression;immunohistochemistry to detect the level of AQP5 protein expression in the submandibular gland tissue of mice.RESULTS:Compared with the normal group,mice in the model group showed no significant changes in body weight,significantly increased water intake(P<0.05),significantly higher expression of IFN-γand TNF-αin serum of mice(P<0.05),and significantly lower expression of IL-10(P<0.05);significantly lower expression of mRNA and protein of AQP5 and M3R(P<0.05);compared with the model group,the western blot and the ZYRZ Pill in high school and the western blot were detected in the submaxillary gland tissues of mice;immunohistochemical method was used to detect the expression level of AQP5 in the submaxillary gland tissues.Compared with the model group,mice in the western medicine group and the high school dose group of ZYRZ consumed significantly less water(P<0.05),the expression of IFN-γand TNF-αin serum was significantly reduced(P<0.05),the expression of IL-10 was significantly elevated(P<0.05),and the expression of AQP5,M3R mRNA,and protein was significantly elevated(P<0.05).CONCLUSION:ZYRZ pill can reduce the inflammatory reaction of submandibular gland in mice with Sj?gren's syndrome model and improve the secretion function of submandibular gland.
