摘要:目的观察加味黑布膏对大鼠痤疮模型的疗效并初步探讨其治疗增生性痤疮瘢痕的作用机制。方法60只Wistar大鼠随机分为空白组、模型组、阳性对照组(维A酸乳膏组)及加味黑布膏低、中、高剂量组,每组10只。除空白组外,其余5组建立大鼠耳廓复合痤疮模型。造模成功后,除空白组和模型组,其余4组分别给药治疗28 d。28 d后,观察6组大鼠耳廓造模区域组织病理变化并检测局部组织中bFGF和IL-17的含量。结果加味黑布膏可不同程度改善大鼠耳廓痤疮模型表皮突延长及角化、减轻炎性细胞浸润及细胞间水肿、抑制成纤维细胞的增长。其中加味黑布膏高剂量组皮肤角质层接近正常;各治疗组大鼠局部组织中bFGF和IL-17的含量均低于模型组(P<0.05);低、中剂量组与阳性对照组比较差异无统计学意义(P>0.05),高剂量组与阳性对照组比较差异有统计学意义(P<0.05)。结论加味黑布膏可降低大鼠耳廓复合痤疮模型局部组织bFGF和IL-17水平,治疗增生性痤疮瘢痕形成的作用机制可能与减少bFGF和IL-17的产生有关。
Objective To determine the effect of modified Heibu ointment in acne model in rats and to explore its mechanism in treating hypertrophic acne scar.Methods Sixty Wistar rats were randomly divided into blank group,model group,positive control group(vitamin A cream group)and enriched Heibu cream low,medium and high dose group,with 10 rats in each group.In addition to the blank group,the other 5 groups were used to establish the model of compound acne.After successful modeling,except the blank group and the model group,the other four groups were given drugs for 28 days.After 28 days,the histopathological changes in auricle modeling area of the 6 groups of rats were observed and the contents of bFGF and il-17 in local tissues were detected.Results Modified Heibu ointment prolonged and improved keratinization of epidermal cells,reduced inflammatory cell infiltration and intercellular edema,and inhibited the growth of fibroblasts in rat auricular acne model.Moreover,the cuticle of the group treated with a high-dose of modified Heibu Ointment was close to normal.The levels of bFGF and IL-17 in the local tissues of rats were significantly lower in each treatment group than they were in the model group(P<0.05).There was a significant difference between the high-dose group and the positive control group(P<0.05).There was no statistical difference between the lowdose group,the middle-dose group and the positive control group(P>0.05).Conclusion Modified Heibu ointment can improve acne scars and reduce the levels of bFGF and IL-17 in local tissues of rat acne model,a mechanism that may be of importance in the treatment of hypertrophic acne scar formation.
