摘要:目的:采用单因素实验和响应面法优化当归叶总黄酮的回流提取工艺,为当归叶中黄酮类化合物的开发利用提供理论依据。方法:采用单因素实验与Box-Behnken中心组合实验相结合的方法,以超声时间、液料比、乙醇体积分数为考察指标,以总黄酮含量为评价指标,优化当归叶总黄酮回流提取工艺。结果:当归叶总黄酮最佳提取工艺条件为乙醇体积分数40%,液料比40 mL/g,提取时间1.5 h,测得总黄酮含量为91.5685 mg/g。试验结果与模型预测值相符。结论:此实验结果为当归叶总黄酮的综合开发利用提供参考依据。
Objective:To optimize reflux extraction process of total flavonoids from leaves of Angelica Sinensis( Oliv.) Diels by using single factor experiment and response surface methodology so as to provide the theoretical evidence for the exploitable and utilization of flavonoids in leaves of Angelica Sinensis( Oliv.) Diels. Methods:Using the single factor experiment combined with Box-Behnken central composite experiment,reflux extracting time,liquid to material ratio and ethanol volume fraction were as the indexes and the content of total flavonoids as the evaluation index,we optimized reflux extraction of total flavonoids from leaves of Angelica Sinensis( Oliv.) Diels. Results:The optimum extraction process was 40% ethanol solution( V/V),with liquid to material ratio being 40 mL/g for 1. 5 hours. The actual yield of total flavonoids was 91. 5685 mg/g,very close to the predictive values. Conclusion:The experimental results for total flavonoids from leaves of Angelica Sinensis( Oliv.) Diels provided a reference for comprehensive exploitation and utilization.
