摘要:目的:观察扶正抑瘤方对胶质瘤(C6)微环境下骨髓间充质干细胞(BMSCs)的影响及作用机制。方法:扶正抑瘤方连续灌胃大鼠并提取大鼠含药血清,利用Transwell小室建立C6与BMSCs共培养环境,BMSCs分成4组进行干预。第1组:BMSCs空白对照组;第2组:BMSCs+扶正抑瘤方含药血清;第3组:BMSCs+C6细胞;第4组:BMSCs+C6细胞+扶正抑瘤方含药血清。通过四甲基偶氮唑蓝(MTT)法检测BMSCs的增殖情况,PCR法检测BMSCs的端粒酶活性,蛋白免疫印迹(Western blot)检测BMSCs中P53蛋白表达。结果:共培养第4组BMSCs的增殖和端粒酶活性比第3组受到较强的抑制(P<0.05);但第4组BMSCs的P53蛋白表达比第3组增高(P<0.05);第1组和第2组BMSCs的增殖、端粒酶活性和P53蛋白的表达均无明显差异(P>0.05)。结论:扶正抑瘤方含药血清对胶质瘤环境下的BMSCs增殖具有抑制作用,其机制可能与抑制BMSCs的端粒酶活性并增加P53表达有关,对BMSCs本身没有影响。
Objective: To observe the effects of Fuzheng Yiliu Fang (FZYLF) on bone marrow mesenchy- mal stem ceils (BMSCs) in C6 micro - environment and explore the mechanism. Methods: Serum containing Fuzheng Yiliu Fang was prepared by ig FZYLF for 3 days (6 times), (:6 and BMSCs cocuhured micro - environ- ment was built by using Transwell chamber, experiment was divided in 4 groups, 1 ) BMSCs (control) group, 2) BMSCs + serum containing FZYLF, 3 ) BMSCs + C6 cells,4)BMSCs + (:6 ceils + serum containing FZYLF. The proliferation of BMSCs was detected by MTY method, telomerase activity of BMSCs was determined by trap - PCR technique ,P53 protein expression in BMSCs was tested by Western blot. Results: The proliferation of BM- SCs and telomerase activity were inhibited obviously in the fourth group ( BMSCs + C6 ceils + serum containing FZYLF)to compare with the third group( BMSCs + C6 cells) ,but the expression of P53 protein in BMSCs was enhanced in the fourth group and that was higher than that of the third group ( P 〈 0. 05 ). All these ( prolifera- tion of BMSCs, telomerase activity and 1〉53 protein expression)were no significant changes in the first (BMSCs) and the second (BMSCs + serum containing FZYLF) group, there were no significant differences between the two groups (P 〉 0. 05 ). Conclusion: Serum containing FZYLF can inhibit the proliferation of BMSCs in C6 micro - environment, the mechanism is related to inhibiting telomerase activity and enhancing P53 expression.
