摘要:目的:研究中风康复膏(PSR)对脑缺血再灌注损伤(CIRI)模型大鼠中炎症反应及氧化应激损伤的影响,并探讨其治疗机制。方法:将50只SPF级SD雄性大鼠随机分为假手术组、模型组、中风康复膏高剂量组(PSR-H组)、中剂量组(PSR-M组)、低剂量组(PSR-L组),除假手术组外其余组大鼠皆采用线栓法制作大脑中动脉缺血再灌注模型,造模成功后各组分别对应给予PSR或生理盐水灌胃干预。采用Zea-Longa's标准评分判断大鼠的神经功能情况并测定脑含水量,TTC染色法观察脑梗死面积,用分光光度法检测TNF-α、IL-1β、ICAM-1、VCAM-1炎症因子及MDA、NOS氧化应激损伤指标。结果:与假手术组相比,模型组的神经功能评分、脑含水量、脑梗死面积和TNF-α、IL-1β、ICAM-1、VCAM-1的含量均显著升高(P<0.01),而PSR各给药组神经功能评分、脑含水量、脑梗死面积和炎症因子的表达量均有所下降,其中PSR-H组、PSR-M组下降较为显著(P<0.01)。氧化应激指标中,相对于假手术组,模型组MDA含量和NOS活力明显升高(P<0.01),PSR给药组MDA含量和NOS活力均有所下降,PSR-H组和PSR-M组下降更为明显(P<0.01)。结论:中风康复膏能够改善脑缺血再灌注损伤大鼠神经功能,具有一定保护作用,其机制可能与改善脑部炎症反应和氧化应激水平有关。
Objective To study the effects of Paste of Stroke Recovery(PSR)on inflammatory response and oxidative stress injury in rats with cerebral ischemia reperfusion injury(CIRI)model and to investigate its therapeutic mechanism.Methods Fifty SPF male rats were randomly divided into sham-operation group,model group,Paste of Stroke Recovery high-dose group(PSR-H Group),middle-dose group(PSR-M Group)and low-dose group(PSR-L group),the model of middle cerebral artery ischemia-reperfusion was made in all groups except sham-operation group,which were treated with PSR or normal saline respectively.The neurological function of rats was evaluated by Zea-Longa’s standard score and the brain water content was measured,TTC staining was used to observe cerebral infarct area,The indexes of TNF-α,IL-1β,ICAM-1,VCAM-1 inflammatory factors and MDA,NOS oxidative stress were measured by Spectrophotometer.Results Compared with sham-operated group,the neurological function score,brain water content,cerebral infarct area and the contents of inflammatory factors TNF-α,IL-1β,ICAM-1 and VCAM-1 in model group were significantly increased(P<0.01),while neurological function score,brain water content,cerebral infarct area and expression of inflammatory factors decreased in each PSR administration group,with more significant decreases in the PSR-H and PSR-M groups(P<0.01).Meanwhile,in oxidative stress indexes,MDA content and NOS activity were significantly higher in the model group relative to the sham-operated group(P<0.01),the level of MDA and the activity of NOS in the PSR group were decreased,especially in PSR-H group and PSR-M group(P<0.01).Conclusions Paste of Stroke Recovery can improve the nerve function of rats with cerebral ischemia-reperfusion injury,and has some protective effects,which may be related to the improvement of brain inflammation and oxidative stress.
