摘要:目的建立一测多评法同时测定皂角刺中原儿茶酸、对羟基苯甲酸、香草酸、丁香酸、表儿茶素、异荭草苷、牡荆苷、异牡荆苷、花旗松素、槲皮苷、木犀草素、槲皮素的含量。方法采用SEVATM C18 Opal(250 mm×4.6 mm,5μm)色谱柱;乙腈-0.1%甲酸溶液为流动相,梯度洗脱;流速为0.6 mL/min;柱温为32℃;检测波长为260 nm、230 nm。以花旗松素为内参物,计算其它11种成分的相对校正因子,采用一测多评法与外标法分别测定皂角刺中12种成分的含量,并对测定结果进行比较。结果原儿茶酸、对羟基苯甲酸、香草酸、丁香酸、表儿茶素、异荭草苷、牡荆苷、异牡荆苷、槲皮苷、木犀草素、槲皮素的相对校正因子分别为1.104、0.777、1.166、2.084、1.378、1.670、2.269、1.803、1.399、1.258、1.142。一测多评法与外标法测得含量值无显著性差异(P>0.05)。结论所建立的一测多评法结果准确、可行性高,可用于皂角刺的质量控制。
Objective To establish a quantitative analysis of multi-components by single-marker method(QAMS)for simultaneous determination of protocatechuic acid,p-hydroxybenzoic acid,vanillic acid,syringic acid,epicatechin,isoorientin,vitexin,isovitexin,taxifolin,quercetin,luteolin and quercetin in Gleditsia sinensis Lam..Methods SEVA C_(18)column(250 mm×4.6 mm,5μm)was used.The mobile phase was acetonitrile-0.1%formic acid solution.The flow rate was 0.6 mL/min,column temperature was 32℃and the detection wavelengths were 260 nm and 230 nm.Taxifolin was chosed as internal reference substance,the relative correction factors(RCF)of the other 11 components were calculated.The contents of 12 components in Gleditsia sinensis Lam.were determined by QAMS and external standard method(ESM)and compared with eachother.Results The RCF of protocatechuic acid,p-hydroxybenzoic acid,vanillic acid,syringic acid,epicatechin,isoorientin,vitexin,isovitexin,quercitrin,luteolin and quercetin were 1.104,0.777,1.166,2.084,1.378,1.670,2.269,1.803,1.399,1.258 and 1.142,respectively.There was no significant difference between the content values measured by the QAMS and ESM(P>0.05).Conclusion The QAMS method established is accurate and feasible,and can be used for the quality control of Gleditsia sinensis Lam.
