摘要:目的研究四君子汤对脾气虚证大鼠运化功能以及学习记忆功能的影响以及分子机制。方法采用综合造模法复制脾气虚证模型。按照随机数字表法将大鼠分为6组:正常组、模型组、对照组和高、中、低3个剂量实验组,每组12只。正常组与模型组给予生理盐水灌胃;对照组给予补中益气汤1.1 g·kg^(-1)灌胃;高、中、低3个剂量实验组分别灌胃四君子汤24,12,6 g·kg^(-1),1次/天,连续21 d。处死前,用Morris水迷宫实验测定大鼠学习记忆能力;处死后,取大鼠血清、海马及小肠组织。用酶联免疫吸附法检测血清中D-木糖含量,用蛋白质印迹法检测大鼠海马、小肠组织中Ghrelin蛋白表达水平。结果正常组、模型组、对照组和高、中、低3个剂量实验组血清D-木糖含量分别为(157.36±18.41),(103.68±22.95),(139.61±18.34),(131.59±18.70),(127.30±6.66)和(125.66±18.81)pg·mg^(-1);这6组小肠组织Ghrelin蛋白表达水平分别为1.25±0.09,0.65±0.01,0.74±0.04,0.98±0.04,0.93±0.07和0.75±0.05;这6组海马组织Ghrelin蛋白表达水平分别为1.12±0.22,0.58±0.02,0.89±0.14,1.10±0.24,0.93±0.13和0.89±0.16。上述这3个指标:模型组与正常组比较,差异均有统计学意义(均P<0.05);对照组和高、中2个剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论四君子汤可明显改善脾虚证大鼠脑肠学习记忆以及消化吸收功能,其作用与上调海马和小肠区域Ghrelin蛋白表达水平有关。
Objective To study the effect of Sijunzi Decoction on the transport function, learning and memory function of rats with Spleen Qi Deficiency syndrome and its molecular mechanism. Methods The spleen Qi deficiency syndrome model was reproduced by comprehensive modeling method. The rats were randomly divided into 6 groups according to random table number: normal group, model group, control group and low, medium and high dose experimental groups, with 12 rats in each group. Rats in normal group and model group were orally administered with normal saline, rats in control group were orally treated with Buzhong Yiqi Decoction(1.1 g·kg^(-1)),while rats in experimental groups were orally administered with Sijunzi Decoction at the dose of 24, 12 and 6 g·kg^(-1), respectively. All the drugs were administered for consecutive 21 d,1 times a day. Morris water maze test was used tomeasure the learning and memory ability of rats before death. The serum,hippocampus and small intestine of rats were taken after death. Enzyme linked immunosorbent assay was used to detect the content of D-xylose in serum,and Western blot was used to detect the expression of ghrelin protein in hippocampus and small intestine. Results The content of D-xylose in normal group,model group,control group,and the three dose of experimental groups were( 157. 36 ± 18. 41),( 103. 68 ± 22. 95),( 139. 61 ± 18. 34),( 131. 59 ± 18. 70),( 127. 30 ± 6. 66) and( 125. 66 ± 18. 81) pg·mg^(-1),respectively;the expression level of ghrelin protein in the small intestine in the 6 groups were 1. 25 ± 0. 09,0. 65 ± 0. 01,0. 74 ± 0. 04,0. 98 ± 0. 04,0. 93 ± 0. 07 and 0. 75 ± 0. 05,respectively;the expression level of ghrelin protein in the hippocampus in the 6 groups were 1. 12 ± 0. 22,0. 58 ± 0. 02,0. 89 ± 0. 14,1. 10 ± 0. 24,0. 93 ± 0. 13 and 0. 89 ± 0. 16,respectively. The above three indicators: comparison between model group and normal group,the differences were statistically significant( all P < 0. 05);comparison between control group,high and medium dose experimental groups,the differences were statistically significant( all P < 0. 05). Conclusion Sijunzi Decoction can significantly improve the learning and memory ability and digestion and absorption function of brain and intestine in rats with spleen deficiency syndrome,which is related to the up regulation of ghrelin protein expression in hippocampus and small intestine.
