摘要:心肌梗死(MI)是临床常见的一种心血管系统疾病,是造成心血管病死亡的主要原因之一,其发病机制复杂,与氧化应激反应相关。核转录因子E_(2)相关因子2(Nrf2)是调节氧化应激反应的关键因子,可调控其下游血红素氧合酶-1(HO-1)的表达,在机体中发挥维持氧化还原稳态的作用。MI的病程中Nrf2与HO-1的生物活性及含量水平均下降,组织抗氧化和抗炎能力减弱,心肌血管内皮受损,血管细胞黏附因子释放,血管内皮功能发生障碍。近年来,诸多基础研究探索中医药通过调控Nrf2/HO-1信号通路治疗MI的作用及机制,结果表明Nrf2/HO-1信号通路是中医药治疗MI的重要潜在靶点。该文就Nrf2/HO-1信号通路在MI中的作用机制及靶向调控该通路的中医药研究进展进行了综述,以期为MI疾病的防治以及进一步的药物开发提供理论依据。
Myocardial infarction(MI)is a common cardiovascular disease in clinical practice.It is one of the main causes of cardiovascular death.Its pathogenesis is complex and related to oxidative stress.Nuclear factor E_(2)-related factor 2(Nrf2)is a key factor regulating oxidative stress response,which can regulate the expression of its downstream heme oxygenase-1(HO-1).It plays a role in maintaining REDOX homeostasis in the body.In the course of MI,the biological activities and content levels of Nrf2 and HO-1 are decreased,the antioxidant and antiinflammatory ability of tissue is weakened,the myocardial vascular endothelium is damaged,vascular cell adhesion factor is released,and vascular endothelial function is impaired.In recent years,many basic studies have explored the role and mechanism of traditional Chinese medicine in the treatment of MI by regulating the Nrf2/HO-1signaling pathway,and the results show that the Nrf2/HO-1 signaling pathway is an important potential target of traditional Chinese medicine in the treatment of MI.This article reviews the mechanism of Nrf2/HO-1 signaling pathway in MI and the research progress of traditional Chinese medicine targeting the regulation of this pathway,in order to provide a theoretical basis for the prevention and treatment of MI and further drug development.
