摘要:目的探讨红芪多糖(HPS)对糖尿病肾病(DN)db/db小鼠肾脏的保护机制。方法以12周龄雄性db/m小鼠10只作为正常组,同周龄雄性db/db小鼠50只随机分为HPS高、中、低剂量组和依那普利组、模型组,每组10只。HPS高、中、低剂量组分别给予HPS 400、200和100mg/(kg·d)灌胃;依那普利组给予依那普利10mg/(kg·d)灌胃;模型组、正常组给予等量生理盐水灌胃;各组均连续灌胃8周。采用免疫组织化学法和免疫印迹法检测各组小鼠肾组织血管内皮生长因子(VEGF)与色素上皮细胞衍生因子(PEDF)蛋白表达;RT-PCR法检测各组小鼠肾组织VEGF与PEDF mRNA表达。结果模型组较正常组VEGF mRNA及蛋白表达均上调(P<0.01),PEDF mRNA及蛋白表达均下调(P<0.01);与模型组比较,除HPS低剂量组外(P>0.05),其余各给药组VEGF mRNA及蛋白表达均下降(P<0.05或P<0.01),PEDF mRNA及蛋白表达均上升(P<0.05或P<0.01)。结论 HPS可通是过调节db/db小鼠肾组织中VEGF、PEDF蛋白和mRNA的表达,从而起到保护DN小鼠肾脏的作用。
Objective To research the protective neys of db/db mice with diabetic nephropathy (DN). mechanism of hedysarum polybotys saccharide (HPS) for kid- Methods Ten 12-week-old male db/m mice were selected as the blank group. Fifty 12-week-old male db/db mice were randomized into the high-dose, middle-dose and low- dose HPS groups, enalapril group and model group, with 10 in each. Each group was given intragastric administration for 8 weeks. The high-dose, middle-dose and low-dose HPS groups were garaged with 400 mg/ (kg d) , 200 mg/ (kg d) and 100mg/ (kg d) HPS respectively. The enalapril group was garaged with 10mg/ (kg d) enala- pril. The model group and blank group were garaged with same volume of normal saline. The expressions of vascular endothelial growth factor (VEGF) and pigment epithelium-derived factor (PEDF) were detected with immunohisto- chemical staining and Western blotting. The expressions of VEGF mRNA and PEDF mRNA were detected with RT- PCR. Results Comparing with the blank group, the expression of VEGF mRNA was significantly increased but the expression of PEDF mRNA was significantly decreased in the model group ( P 〈 0.01 ). Comparing with the model group, the expressions of VEGF and VEGF mRNA were significantly decreased but the expressions of PEDF and PEDF mRNA were significantly increased in all administration groups ( P 〈 0. 05 ) except the low-dose HPS group. Conclusion HPD can protect the kidneys of DN mice by regulating the expressions of VEGF, PEDF, VEGF mRNA and PEDF mRNA in renal tissue of db/db mice.
