摘要:目的运用中心组合设计-响应面法对中药复方苦柏坐浴颗粒剂(简称洗剂)的提取工艺进行综合优化。方法在单因素实验基础上,采用中心组合设计-响应面法考察乙醇浓度、料液比及提取时间对洗剂中苦参碱、氧化苦参碱、总生物碱及浸膏提取效率的影响。结果最佳提取工艺为回流提取法,提取溶剂为50%乙醇溶液,料液比为1∶20,提取时间为90 min。结论优化后的提取工艺操作简便,稳定可行,能较大程度提高该洗剂的提取效率。
Objective To optimize the extraction process of compound Kubai sitz bath granules by the central composite design-response surface methodology. Methods On the basis of single factor tests,the effect of the ethanol concentration,solid-liquid ratio and extracting time on the extraction of matrine,oxymatrine,total alkaloids and total extract were investigated by the central composite design-response surface methodology. Results The conditions for the optimal extraction process were as follows:the reflux extraction with 50% aqueous ethanol,the 1∶20 of solid-liquid ratio of the raw material to the 50% aqueous ethanol solution,and the90 minutes of refluxing time. Conclusion The optimized extraction process was easily operable,stable and feasible,which could effectively improve the extraction efficiency of the extraction process of compound Kubai sitz bath grannules.
