摘要:目的观察白及多糖对胃溃疡模型大鼠血清白细胞介素(IL)-2R、IL-4含量及胃组织Caspase-8表达的影响,探讨其治疗胃溃疡的作用机制。方法60只Wistar大鼠随机分为空白组和造模组,造模大鼠采用乙酸直接烧灼法制作胃溃疡模型,将成模大鼠按随机数字表法分为模型组、阳性对照组和白及多糖高、中、低剂量组,每组10只。空白组和模型组大鼠给予10 mL/(kg·d)蒸馏水灌胃,白及多糖高、中、低剂量组大鼠分别给予白及多糖0.5、0.25、0.125 g/(kg·d)药液灌胃,阳性对照组大鼠给予雷尼替丁0.3 g/(kg·d)药液灌胃,连续15 d。ELISA检测大鼠血清IL-2R、IL-4含量,实时荧光定量PCR检测大鼠胃组织IL-2R、IL-4及Caspase-8基因表达,Western blot检测Caspase-8蛋白表达。结果与空白组比较,胃溃疡大鼠一般生存状况较差,IL-2R含量及基因表达明显升高、IL-4含量及基因表达明显降低(P<0.05,P<0.01),胃组织Caspase-8基因和蛋白表达显著升高(P<0.05);与模型组比较,白及多糖高、中、低剂量组大鼠IL-2R含量及基因表达均降低、IL-4含量及基因表达均升高,Caspase-8基因和蛋白表达均降低,白及多糖高剂量组差异有统计学意义(P<0.05)。结论白及多糖通过下调模型大鼠血清IL-2R、上调IL-4含量及下调胃组织Caspase-8表达保护胃黏膜,达到治疗胃溃疡的作用。
Objective To observe the effects of Bletilla baicalensis polysaccharides on serum IL-2R,IL-4 level and Caspase-8 expression in model rats with gastric ulcer.Methods Totally sixty Wistar rats were randomly divided into blank group and modeling group.The gastric ulcer model was reproduced by acetic acid direct cauterization.The model rats were divided into model group,positive control group and Bletilla baicalensis polysaccharide high-,medium-,and low-dosage groups according to random number table method,with 10 rats in each group.Rats in blank group and model group were given 10 mL/(kg·d)distilled water for gavage;rats in Bletilla baicalensis polysaccharide high-,medium-,and low-dosage groups were given 0.5,0.25,0.125 g/kg Bletilla baicalensis polysaccharide for gavage;rats in positive control group were given 0.3 g/(kg·d)ranitidine for gavage.All groups received medicine for successive 15 d.After 15 days of treatment,serum IL-2R and IL-4 levels were detected by ELISA,and real-time fluorescence quantitative PCR was used to detect the gene expressions of IL-2R,IL-4 and Caspase-8 in stomach tissues.The protein expression of Caspase-8 was detected by Western blot.Results Compared with the blank group,gastric ulcer rats had worse general survival,higher IL-2R content and gene expression level,lower IL-4 content and gene expression level(P<0.05,P<0.01),and higher levels of Caspase-8 gene and protein expression in stomach tissue(P<0.05).Compared with the model group,the levels of IL-2R and gene expression in Bletilla baicalensis polysaccharide high-,medium-,and low-dosage groups decreased;the levels of IL-4 and gene expression increased;the levels of Caspase-8 gene and protein expression decreased,with statistical significance in Bletilla baicalensis polysaccharide high-dosage group(P<0.05).Conclusion Bletilla baicalensis polysaccharide can protect gastric mucosa to treat gastric ulcer by down-regulating serum IL-2R,up-regulating IL-4 content and down-regulating Caspase-8 expression in model rats.
