摘要：目的观察黄金胶囊对2型糖尿病(T2DM)大鼠肝组织eIF2α、GRP78表达的影响,探讨其可能的作用机制。方法采用高糖高脂饲料喂养联合小剂量链脲佐菌素腹腔注射制作T2DM大鼠模型。成模大鼠随机分为模型组、罗格列酮组和黄金胶囊低、中、高剂量组,另取8只大鼠为正常组。各给药组给予相应药物灌胃,模型组和正常组给予生理盐水灌胃,连续10周。Western blot和RT-PCR分别检测大鼠肝组织eIF2α、GRP78蛋白和mRNA的表达,HE染色观察肝组织病理形态。结果与正常组比较,模型组大鼠eIF2α、GRP78蛋白和mRNA表达均明显升高(P<0.05);与模型组比较,各给药组大鼠eIF2αmRNA表达明显降低(P<0.05),罗格列酮组较黄金胶囊中、低剂量组差异有统计学意义(P<0.05),黄金胶囊高剂量组与罗格列酮组作用相当(P>0.05)。HE染色显示黄金胶囊各剂量组大鼠肝脏病理损害减轻,高剂量组减轻最明显。结论黄金胶囊可抑制模型大鼠肝组织eIF2α、GRP78表达的上调,减轻病理损害,从而改善肝组织内质网应激,降低血糖。
Objective To explore the effects of Huangjin Capsules on the expressions of eIF2 and GRP78 in the liver of type 2 diabetes mellitusrats(T2DM);To explore its possible mechanism of action.Methods T2DM rat model was established by high-sugar and high-fat diet combined with intraperitoneal injection of low-dose streptozotocin.The model rats were randomly divided into model group,rosiglitazone group,Huangjin Capsules low-,medium-and high-dosage groups.Another 8 rats were chosen as normal control group.Each administration group was given relevant medicine for gavage,while model group and normal control group were given saline for gavage,for consecutive 10 weeks.Western blot and RT-PCR were used to detect theprotein and mRNA expressions of eIF2αand GRP78 in rat liver tissue.HE staining was used to observe the pathological morphology of liver tissue.Results Compared with the normal control group,the protein and mRNA expressions of EIF2 and GRP78 significantly increased in the model group(P<0.05).Compared with model group,eIF2 mRNA expressions decreased in both rosiglitazone group and Huangjin Capsules medium-and low-dosage groups,with statistical significance(P<0.05).The effects of Huangjin Capsules high-dosage group were the same as rosiglitazone group.HE staining showed that the liver pathological damage was alleviated in the rats in Huangjin Capsules groups,and the most significant reduction was observed in the high-dosage group.Conclusion Huangjin Capsules can inhibit the up-regulation of expressions of eIF2 and GRP78 in liver tissue of model rats and reduce pathological damage,so as to improve the liver endoplasmic reticulum stress and reduce blood glucose.
