摘要:目的观察参白合剂对溃疡性结肠炎(UC)小鼠疾病活动指数(DAI)及血清中超氧化物歧化酶(SOD)和丙二醛(MDA)的影响及其作用机制。方法将50只小鼠随机分为空白组、模型组、柳氮磺胺吡啶(SASP)组、参白合剂高剂量组(参高组)、参白合剂低剂量组(参低组)5组,每组10只。造模各组自由饮用3%葡聚糖硫酸钠溶液,连续10 d。第11日始各组灌肠干预治疗,其中空白组、模型组给予生理盐水0.2 mL/10 g,SASP组给予0.025 g/mL SASP混悬液,参高组、参低组分别给予23.5、11.75 g/mL参白合剂混悬液,连续10 d。在第1、10、20日对小鼠进行DAI评分。第21日采用眼球取血法取血约2 mL,ELISA法检测SOD和MDA含量。结果 DAI评分显示:造模第10日,各造模组与空白组比较差异有统计学意义(P<0.05);实验第20日,各治疗组与模型组比较差异有统计学意义(P<0.05)。模型组SOD浓度低于空白组(P<0.05),MDA浓度高于空白组(P<0.05);SASP组、参高组、参低组SOD浓度明显高于模型组(P<0.05),MDA浓度明显低于模型组(P<0.05);SASP组与参高组比较差异无统计学意义(P>0.05)。结论参白合剂通过提高SOD含量、减少脂质过氧化物生成,从而干预UC的病理过程。
Objective To observe the effect and mechanism of Shenbaiheji on the disease activity index(DAI),serum SOD and MDA of ulcerative colitis(UC) mice.Methods Fifty mice were randomly divided into blank group,the model group,SASP group,and Shenbaiheji high dose group and low dose group(10 mice for each group).Except blank group,others freely drank 3% DSS solution for 10 days.On 11th day,blank group and model group were treated by enema with NS 0.2 mL/10 g,SASP group with 0.025 g/mL SASP suspension liquid,high and low dose group with 23.50,11.75 g/mL Shenbaiheji.On the first,10th and 20th day,the DAI was evaluated.On 21st day,blood was taken from mice eyeball 2 mL to detect the content of MDA and SOD with ELISA method.Results DAI of the model group had statistically significant difference with blank group(P0.05) on the10th day,and the treated groups had statistically significant difference with model group(P0.05) on the 20th day.SOD concentration of model group was lower than blank group(P0.05),MDA was higher than the blank group(P0.05).SOD concentration of SASP group and Shenbaiheji high,low dose group was obviously higher than that of the model group(P0.05),MDA concentration was significantly lower than the model group(P0.05).SASP group had no statistically significant diffence with Shenbaiheji high dose group(P0.05).Conclusion Shenbaiheji can intervent UC by increasing SOD and decreasing LPO.
