摘要:目的研究电针刺对去卵巢骨质疏松模型大鼠全身骨密度(BMD)和骨矿物含量(BMC)及其血清对体外培养成骨细胞的骨形成因子骨钙素(OPG)和骨吸收因子(RANKL)mRNA表达的影响。方法选取SPF级2月龄SD雌性大鼠48只,随机分为药物组、针刺组、模型组和空白组,每组12只。其中空白组不做任何处理,其余36只采用手术方法切除双侧卵巢。各组大鼠于SPF级实验室饲养3个月后,双能x射线骨密度仪检测全身BMD和BMC,确定造模成功。药物组大鼠灌胃服用戊酸雌二醇3个疗程,针刺组大鼠采用针刺后通电治疗3个疗程并服用药物组相同体积的蒸馏水,模型组服用相应体积的蒸馏水,空白组不做任何刺激,常规饲养。随后,水合氯醛麻醉各组大鼠,心脏采血,各组血液分别经滤网过滤、56℃水浴处理后加入体外培养的大鼠成骨细胞培养液中,处理3天后多聚甲醛固定成骨细胞,采用原位杂交法检测成骨细胞内OPG、RANKL mRNA的表达量。结果与空白组相比,模型组大鼠全身BMD,BMC显著降低,服用药物或针刺处理后,BMD和BMC显著增高,与模型组相比具有显著性差异(P<0.01)。各组大鼠血清处理成骨细胞后,空白组骨形成指标OPG mRNA表达量最高,模型组最低,药物组和针刺组OPG mRNA表达量显著高于模型组(P<0.01)。此外,空白组骨吸收指标RANKL mRNA表达量最低,模型组最高,药物组和针刺组RANKL mRNA表达量显著低于模型组(P<0.01)。结论针刺能有效升高大鼠全身BMD和BMC,同时针刺处理后的大鼠血清具有很好的促进骨形成和抑制骨吸收的能力。
Objective To observe the effects of electroacupuncture on bone mineral density and bone mineral content in ovariectomy rats and the effects of serum from acupunctured rats on the expression of OPG and RANKL in osteoblasts cultured in vitro. Method Forty-eight female two-month old SD rats from the grade of SPF were selected and divided randomly into four groups,in which three groups undergone the surgery of bilateral ovariectomy and then named the groups of electroacupuncture,the group of western medicine and the group of model. The other group were blank group which did not receive any procedure. Rats in the western medicine group were administrated estradiol valerate by means of intragastrical administration for three courses. Rats in the electroacupuncture group were treated by acupuncture and received the same volume distilled water for three months. Rats in the model group were administrated with distilled water for three months. Rats in the blank group were not treated. After three months when the rats were raised in the laboratory of SPF by the common way,they were assessed and the result showed that the models were established. Then the all groups of rats were anesthetized by the chloral hydrate and hemospasia from heart was performed.Blood samples of each group undergone filter mesh and water bath,and the serum was added to osteoblasts cultured in vitro. After three days osteoblasts were fixed using paraformaldehyde. The mRNA of OPG and RANKL were detected by means of immunofluorescence cytochemistry. Results Compared with the blank group,the group of model had significantly lower bonemineral density and bone mineral content. After the treatment of western medicine or electroacupuncture,they were increased,and the differences were significant( P〈0. 01) compared with the group of model. After the osteoblasts were cultured in the serum,the blank group was the highest,the model group was the lowest and lower than the western group and the electroacupuncture group on the mRNA of OPG( P〈0. 01). The model group was the highest on the mRNA of RANKL,but after the treatment,that of the western group and the electroacupuncture group decreased( P〈0. 01). Conclusion The treatment of electroacunpuncture could mostly raise bone mineral density and bone mineral content in rats. After the treatment of electroacupuncture, the serum could increase bone formation and decreased bone resorption.
