摘要:目的探讨扶金化积丸对Lewis肺癌荷瘤模型小鼠的抑瘤作用及机制。方法选取健康C57BL/6小鼠接种Lewis肺癌瘤株进行造模,小鼠随机分成正常组,荷瘤对照组,阳性组(贞芪扶正颗粒5 g/kg),扶金化积丸高、中、低剂量组(剂量分别为1.749、0.875、0.437 g/kg)。各组药物干预后,检测荷瘤小鼠瘤质量、瘤体体积,计算抑瘤率、器官指数。全自动血液分析仪进行血细胞计数及血红蛋白(HGB)水平检测,ELISA检测血清TNF-α、IL-2、IFN-γ水平,HE染色观察瘤组织病理学改变,免疫组化法检测MMP-9、NF-κB表达。结果与荷瘤对照组比较,扶金化积丸干预可使荷瘤小鼠瘤质量、瘤体体积减小(P<0.05,P<0.01),脾脏、胸腺指数及红细胞(RBC)、白细胞(WBC)、血小板(PLT)数目及HGB水平均升高(P<0.05,P<0.01),TNF-α、IFN-γ、IL-2水平均增加(P<0.05,P<0.01),瘤组织核固缩较多,有明显坏死及出血现象,出现细胞凋亡,MMP-9、NF-κB表达均降低(P<0.05,P<0.01)。结论扶金化积丸对Lewis肺癌荷瘤小鼠具有抑瘤作用,其机制可能与提高机体免疫力,降低NF-κB、MMP-9表达有关。
AIM To investigate the antitumor effects of Fujin Huaji Pills on mice bearing Lewis lung cancer tumor, and to explore the underlying mechanisms.METHODS C57 BL/6 mice were used to establish Lewis lung cancer models. The mice were randomly divided into normal control group, tumor-bearing control group, Zhenqi Fuzheng Granules group(5 g/kg), and Fujin Huaji Pills groups for the intervention of high dose, medium dose, and low dose uses of the pills at concentration of 1.749, 0.875, 0.437 g/kg, respectively. After treatment, mice had their tumor weight, tumor volume, tumor-inhibition rate and organ indices levels assessed;theri blood cell counts and hemoglobin(HGB) level determined;their TNF-α, IL-2, and IFN-γ levels detected by ELISA;their histopathological changes in tumor tissues observed by HE staining;and their protein expressions of MMP-9 and NF-κB evaluated by immunohistochemical staining.RESULTS As compared with the tumor-bearing control group, Fujin Huaji Pills groups shared decreased tumor weight and tumor volumn(P<0.05, P<0.01);increased indices levels of the thymus and the spleen(P<0.05), much more counts of red blood cell(RBC), white blood cell(WBC), platelets(PLT), and higher HGB level(P<0.05, P<0.01);enhanced TNF-α, IFN-γ, and IL-2 levels(P<0.05, P<0.01);and more obvious pyknosis, necrosis, and apoptosis in tumor tissues, and reduced expressions of MMP-9 and NF-κB(P<0.01, P<0.01).CONCLUSION Fujin Huaji Pills exert antitumor effect on mice bearing Lewis lung cancer, and the mechanisms may be related to the improved organism immunity and compromised MMP-9 and NF-κB expressions.
