摘要:为了探索党参不定根生长和增殖的适宜条件,切取党参组织培养的不定根接种到培养基中,研究根的部位和接种方式、激素种类和浓度、培养基种类和氮源对不定根生长和增殖的影响。结果表明,单根和根尖诱导不定根增殖较好。不同基本培养基对不定根诱导能力不同,最佳的培养基为1/2 MS和B5。激素种类和浓度为IBA 0.1 mg/L和BA 0.5 mg/L+NAA 0.01 mg/L时诱导培养不定根的效果较好。不同氮源对党参不定根诱导的影响中,以NO_(3)^(-)∶NH_(4)^(+)为2∶1时效果较好。适宜不定根诱导和增殖的培养基为1/2 MS+IBA 0.1 mg/L+2%蔗糖+0.7%琼脂。
In order to explore the suitable conditions for the growth and proliferation of adventitious roots of Codonopsis pilosula(Franch.)Nannf.,the adventitious roots of tissue culture were cut and inoculated into the medium,and the effects of the root location,inoculation method,hormone type and concentration,medium type and nitrogen source on the growth and proliferation of adventitious roots were studied.The results showed that single root and apex induced adventitious root proliferation was better.Different basic medium had different ability to induce adventitial root,and the best medium were 1/2 MS and B 5.When the hormone types and concentrations were 0.1 mg/L for IBA and 0.5 mg/L for BA and 0.01 mg/L for NAA,the effect of induction of adventitious roots was better.Among the effects of different nitrogen sources on induction of advective roots of Codonopsis pilosula(Franch.)Nannf.,and the was best when NO_(3)^(-)∶NH_(4)^(+)was 2∶1.The optimal medium for adventitic root induction and proliferation was 1/2 MS+IBA 0.1 mg/L+2%sucrose+0.7%AGAR.
