摘要:目的比较改良气管穿刺法、常规气管穿刺法及气管切开法注射博莱霉素制作肺间质纤维化大鼠模型的优劣。方法将60只Wistar大鼠随机分为改良气管穿刺组、常规气管穿刺组及气管切开组,各组大鼠麻醉后分别采用相应的给药方法一次性推注博莱霉素5 mg/kg制作肺间质纤维化大鼠模型,造模14,28 d后分批处死大鼠,取大鼠右肺下叶组织分别进行HE染色和Masson染色,然后分别进行肺组织病理损伤评分和肺组织纤维化评分。结果气管切开组大鼠死亡7只,死亡率为35%,改良气管穿刺组和常规气管穿刺组死亡率均为0;改良气管穿刺组比气管切开组操作耗时更少,与常规气管穿刺组比较,差异有高度统计意义(P<0.01);常规气管穿刺组与气管切开组在病理损伤及肺纤维化评分方面比较,差异有统计意义(P<0.05),而改良气管穿刺组与气管切开组比较,差异无统计意义(P>0.05)。结论改良气管穿刺法比常规气管穿刺法造模效果更好,与气管切开插管法相当,但操作更简便,耗时更少,大鼠死亡率更低。
Objective To compare three methods in modeling rats with pulmonary fibrosis induced by intratra-cheal injection of bleomycin. They are improved method of tracheal puncture,conventional method of tracheal punc-ture and tracheotomy.Methods 60 Wistar rats were randomly divided into improved tracheal puncture group,con-ventional tracheal puncture group and tracheotomy group. The rats were given disposable injection of bleomycin(5 mg/kg) by corresponding administration to cause pulmonary interstitial fibrosis when they were anesthetized.The rats were killed in batches on 14 thday and 28 thday. Lung tissues in the lower lobe of models were taken for HEand Masson staining. Pathological injury and fibrosis were scored.Results 7 rats in the tracheotomy group died,the mortality rate was 35%. No rat died in the other two groups. Compared with tracheotomy,the improved methodof tracheal puncture had less time-consuming. Compared with conventional tracheal puncture group,there was asignificant difference(P<0. 01). There was a significant difference between the conventional tracheal puncturegroup and tracheotomy group in comparing pathological injury and pulmonary fibrosis(P<0. 05). However,therewas no difference between the improved tracheal puncture group and tracheotomy group(P>0. 05).Conclusion The improved method of tracheal puncture is better than the conventional method. It is simpler,quicker and moreconvenient than tracheotomy and the mortality in rats is lower.
