摘要:目的考察不同压敏胶基质对小儿清热宣肺贴中有效成分释放速率和微透析累积透过率的影响,测定其释药和透皮性能,为小儿清热宣肺贴优选出最适宜的基质。方法采用HPLC测定0.5~8 h苦杏仁苷的累积溶出量和给药8 h苦杏仁苷微透析累积透过量,比较不同压敏胶基质对小儿清热宣肺贴中有效成分释放速率和透皮吸收的影响。结果小儿清热宣肺凝胶基质贴溶出8 h时苦杏仁苷基本完全释放,中分子量聚异丁烯基质贴溶出8 h时苦杏仁苷溶出度约为10%,高分子量聚异丁烯基质贴在此时间段内未检测到苦杏仁苷的释放。凝胶基质贴给药8 h苦杏仁苷微透析累积透过量为(29.13±3.09)%,中分子量聚异丁烯基质贴在此时间段内微透析累积透过量为0。结论水凝胶基质有利于药物有效成分的释放和透皮吸收,更适合用作小儿清热宣肺贴的基质。
Objective To investigate the effects of different kinds of pressure-sensitive adhesive matrix on release rate of effective components and the cumulative transmittance of microdialysis on pediatric Qingre Xuanfei Plaster;To measure the release and penetration properties;To choose a suitable substrate for pediatric Qingre Xuanfei Plaster.Methods HPLC method was used for determine 0.5–8 h laetrile cumulative dissolution amount and 8 h laetrile cumulative transmittance amount of microdialysis.Effects of different kinds of pressure sensitive adhesive matrixs on release rate of effective components and transdermal absorption on pediatric Qingre Xuanfei Plaster were compared.Results When pediatric Qingre Xuanfei Plaster of hydrogel matrix was dissoluting in 8 h,the laetrile was almost released in full;when plaster of the medium molecular weight polyisobutylene matrix was dissoluting in 8 h,the laetrile release amount was about 10%.When plaster of the high molecular weight polyisobutylene matrix was in this period,the release of laetrile was not detected.When hydrogel matrix was dosing for 8 h,the laetrile cumulative transmittance amount of microdialysis was(29.13±3.09)%;when the medium molecular weight of polyisobutene matrix plaster was in this period,the cumulative transmittance amount of microdialysis was 0.Conclusion Hydrogel matrix is conductive to the release of effective components and transdermal absorption,and is more suitable for being used as matrix in pediatric Qingre Xuanfei Plaster.
