摘要:目的研究花椒炮制前后挥发油有效部位在小鼠体内的抗炎、镇痛作用。方法 (1)镇痛实验:选择疼痛反应合格(痛阈值在5~30 s内)的小鼠并随机分为6组:空白对照组(2%吐温80);高、低2个剂量生品花椒挥发油组(4,2g·kg^(-1));高、低2个剂量清炒品花椒挥发油组(4,2 g·kg^(-1));阳性对照组(去痛片0. 5 g·kg^(-1)),每组10只。灌胃给药,1天/次,连续4 d,用小鼠热板法和醋酸扭体法研究花椒生品及清炒品挥发油部位的镇痛作用(测定痛阈值,计算扭体反应抑制率)。(2)抗炎实验:将昆明种小鼠随机分为6组:空白对照组(2%吐温80);高、低2个剂量生品花椒挥发油组(2,1 g·kg^(-1));高、低2个剂量清炒品花椒挥发油组(2,1 g·kg^(-1));阳性对照组(阿司匹林0. 5 g·kg^(-1)),每组10只。灌胃给药,1天/次,连续7 d。于末次给药后1 h,以二甲苯致小鼠耳肿胀为模型,观察花椒生品与清炒品挥发油部位的抗炎作用(肿胀抑制率)。结果(1)镇痛作用:花椒生品、炮制品挥发油部位对由冰醋酸所引起的疼痛具有明显的缓解效果,高、低2个剂量生品花椒挥发油组和高、低2个剂量清炒品花椒挥发油组与对照组小鼠在20 min内的扭体次数分别为(19. 50±3. 31),(27. 20±3. 91),(18. 40±3. 10),(21. 10±2. 85),(19. 20±3. 79),(49. 60±7. 62)次,4个给药组与空白对照组相比,差异均有统计学意义(均P <0. 01)。(2)抗炎作用:高、低2各剂量生品花椒挥发油组和高、低2个剂量清炒品花椒挥发油组与对照组的耳肿胀值分别为(2. 1±0. 2),(5. 7±0. 3),(1. 7±0. 2),(3. 5±0. 3),(0. 7±0. 3),(6. 0±0. 2) mg,高剂量生品花椒挥发油组和2个剂量清炒品花椒挥发油组与对照组比较,差异均有统计学意义(均P <0. 01)。结论花椒挥发油具有显著的抗炎镇痛活性,且作用与剂量相关;花椒炮制后其挥发油的抗炎、镇痛作用呈增强的趋势。
Objective To study the anti-inflammatory and analgesic effects of Volatile oil of Zanthoxylum bungeanum before and after processing in mice. Methods( 1) Analgesia test: the mice with qualified pain response were randomly divided into six groups: control group( 2%Tween 80);high and low dosage of Volatile oil of Zanthoxylum bungeanum( 4,2 g·kg-1) groups;high and low doses of Volatile oil of fried prickly ash( 4,2 g·kg-1) groups;positive control group( Qutongpian0. 5 g·kg-1),10 rats in each group. Administered by intragastric administration,1 times a day,for 4 d. The analgesic effect of Volatile oil of Zanthoxylum bungeanum L. was study by hot plate method and aceticacid writhing method( the pain threshold was measured and the inhibition rate of writhing reaction was calculated).( 2) Anti-inflammatory test: Kunming mice were randomly divided into six groups: blank control group( 2% Tween80);high and low dosage of Volatile oil of Zanthoxylum bungeanum( 2,1 g·kg-1);high and low doses of Volatile oil of fried prickly ash( 2,1 g·kg-1);positive control group( aspirin 0. 5 g·kg-1),10 rats in each group. Administered by intragastric administration,1 times a day,for 7 d. The anti-inflammatory effect( swelling inhibition rate) of the Volatile oil of Zanthoxylum bungeanum and fried Zanthoxylum bungeanum were observed in mice model of the xylene-induced ear swelling. Results( 1) Analgesic effect: the Volatile oil of Zanthoxylum bungeanum L. had obvious relief effect on the pain caused by acetic acid. The number of writhing after 20 min in Volatile oil-L,-H groups,Volatile oil of fried prickly ash-L,-H groups and control group were( 19. 50 ± 3. 31),( 27. 20 ± 3. 91),( 18. 40 ± 3. 10),( 21. 10 ± 2. 85),( 19. 20 ± 3. 79),( 49. 60 ± 7. 62) times. Comparison between 4 drugs groups and control group,the difference were significantly( all P < 0. 01).( 2) Anti-inflammatory effect: The ear swelling values in the 5 groups were( 2. 1 ± 0. 2),( 5. 7 ± 0. 3),( 1. 7 ± 0. 2),( 3. 5 ± 0. 3),( 0. 7 ± 0. 3),( 6. 0 ± 0. 2) mg;except for the Volatile oil-L group,comparison between the other three drugs groups and control group,the difference were significantly( all P < 0. 01). Conclusion The essential oil of Zanthoxylum bungeanum had a significant anti-inflammatory and analgesic activity,and the effect is related to the dosage. The anti-inflammatory and analgesic effect of the Volatile oil of Zanthoxylum bungeanum is stronger than that of the processed Zanthoxylum bungeanum.
