摘要:目的:观察当归对阿托品诱发小鼠便秘的治疗作用。方法:动物分为对照组、阿托品组、当归低剂量组、当归中剂量组、当归高剂量组,对照组灌胃0.4 m L生理盐水,其余各组以8 mg/kg剂量连续灌胃阿托品4 d,同时以4.8 g/kg、9.6 g/kg、14.4 g/kg剂量连续灌胃当归水煎液4 d,末次给药前12 h停食给水,当归药液、生理盐水均含10%活性炭,给药30 min后,颈椎脱臼处死各组8只小鼠,以炭末推进率、6 h时间内的排粪重量为指标,观察当归对阿托品诱导小鼠肠运动的影响。采用SPSS17.0数据分析软件,应用单因素方差分析SNK进行组间比较。结果:与阿托品组比较,当归中剂量组炭末推进率差异显著(P<0.05),高剂量组差异极显著(P<0.01);与空白组比较,6 h内阿托品组排粪数量、粒数差异不显著;与阿托品组比较,当归中剂量组、当归高剂量组排粪数量、粒数差异不显著,但有促进排粪趋势。结论:当归可以改善阿托品诱发的肠蠕动迟缓,其机制可能为解除阿托品引起的肠肌麻痹。
Objective: To determine the therapeutic effect of Angelica Sinensis(AS) on atropine-induced constipation. Methods:Mice were randomly divided into control, atropine group(8 mg/kg), AS high-dose(14.4 g/kg), AS medium-dose(9.6 g/kg) and AS lowdose(4.8 g/kg) groups with 8 in each. Control group was given 0.4 m L of normal saline by gavage, other groups were given 8.0 mg/kg of atropine and different dose of AS extract with water for 4 days simultaneously. After food supply were prohibited for 12 h, every tested sample mixed 10% powder of carbon is administrated in last ig, 30 min later, partial mice of every group were executed by removing cervical vertebra for observing charcoal-powder propelling rate, and other mice of every group were recorded to their amount of feces in 6 hours after administration. Results: Compared with atropine group, Charcoal-powder propelling rate was increased in AS highdose group(ASHDG, P〈0.01) and AS medium-dose group(ASMDG, P〈0.05). The feces amounts in ASHDG and ASMDG had no statistic significance compared with control group. Conclusion: Radix Angelica Sinensis can resist intestinal hypoperistalsis induced by atropine, the mechanism may be abolishment of intestinal muscle paralysis caused by atropine.
