摘要:目的研究中风康复膏对缺血性脑卒中模型大鼠脑组织中白细胞介素(IL)-17、IL-6含量及神经功能缺损评分的影响,分析中风康复膏治疗缺血性脑卒中的作用机制。方法将40只SPF级SD雄性大鼠采用随机数字表法分为假手术组、模型组、阳性对照组和中风康复膏组4组,每组10只,各组再根据不同时间点分为6,24,72 h 3个亚组,其中6 h 3只、24 h 3只、72 h 4只。除假手术组外,其余3组采用线栓法复制大鼠大脑中动脉栓塞模型。各组分别灌胃相应药物及生理盐水。每组各亚组分别于造模后6,24,72 h进行神经功能缺损评分,并在相应时间点分批处死大鼠取脑组织,采用酶联免疫吸附测定(ELISA)法检测脑组织中的IL-17和IL-6含量。结果与同时间点假手术组比较,模型组大鼠脑组织中IL-17、IL-6含量及神经功能缺损评分在各时间点均明显升高,差异有统计学意义(P<0.01);与同时间点模型组比较,阳性对照组与中风康复膏组大鼠脑组织中IL-17、IL-6含量及神经功能缺损评分在各时间点均明显降低,差异有统计学意义(P<0.01)。结论中风康复膏可通过降低脑组织中的IL-17、IL-6含量抑制炎症反应的发生,从而保护神经细胞,改善神经功能,防治缺血性脑损伤。
Objective To study the effects of Zhongfeng Kangfu Gao(中风康复膏,ZFKFG)on the content of interleukin-17(IL-17)and interleukin-6(IL-6)in brain tissue and neurological deficit score in model rats with ischemic cerebral stroke,and to analyze its mechanism.Methods Forty SPF SD male rats were divided into 4 groups by random number table method,with 10 rats in each group,which were sham operation group,model group,positive control group and ZFKFG group.Each group was further divided into 3 subgroups including 6,24 and 72 hours according to different time points,in which 3 at 6 hours,3 at 24 hours and 4 at 72 hours.Except the sham operation group,the other 3 groups used suture-occluded method to replicate the model rats of middle cerebral artery occlusion.Each group was given intragastric administration of corresponding drugs and normal saline.At 6,24 and72 hours after establishing models,the neurological deficit score was performed in each subgroup,and the rats were killed in batches at corresponding time points and their brain tissue was taken out.The content of IL-17 and IL-6 in brain tissue was detected by enzyme-linked immunosorbent assay(ELISA)method.Results Compared with the sham operation group at the same time point,the content of IL-17 and IL-6 in brain tissue and the neurological deficit scores of rats in the model group significantly increased at each time point,and the differences were statistically significant(P<0.01).Compared with the model group at the same time point,the content of IL-17 and IL-6 in brain tissue and the neurological deficit scores of rats in the positive control group and ZFKFG group significantly decreased at each time point,and the differences were statistically significant(P<0.01).Conclusion ZFKFG can decrease the content of IL-17 and IL-6 in brain tissue,inhibit the inflammatory reactions,thus playing the role of protecting nerve cells,improving nerve function and preventing cerebral ischemia injury.
