摘要:目的:建立抗生素相关性大肠湿热证大鼠模型,从体征、肠道菌群、血清及结肠组织炎症因子、结肠组织病理等进行评价。方法:采用肥甘及辛辣饮食损伤大鼠肠道引起"内湿、内热",利用人工气候箱模拟"外湿、外热"环境,以生物因素和抗生素应激诱导建立抗生素相关性大肠湿热证模型,观测模型大鼠的一般状态、体质量、摄食量、肛温,对肠道敏感性和粪便性状进行评分,测定粪便含水量及血清TNF-α、IL-1β、DAO、D-乳酸含量,检测肠黏膜SIgA及结肠组织MPO含量,计数结肠内容物中乳酸杆菌、双歧杆菌、肠球菌、大肠杆菌,分析结肠组织形态变化。结果:复合因素诱导建立的A-DHS模型大鼠体质量减轻、摄食量减少、肛温升高、肠道敏感性增加、4 h内排便次数增多、粪便性状评分增加、粪便含水量升高,血清TNF-α、IL-1β、DAO和D-乳酸含量均显著升高,结肠组织MPO含量升高,肠黏膜SIgA含量降低,肠内容物中乳酸杆菌、双歧杆菌显著减少,而肠球菌、大肠杆菌显著增加,HPS评分显著升高;175 mg/kg柳氮磺吡啶治疗7 d,各指标均有所恢复。结论:复合因素可成功诱导建立抗生素相关性大肠湿热证大鼠模型,其病理特征与临床症状基本一致。
Objective:To establish and evaluate the rats model of antibiotic related large intestine dampness-heat syndrome induced by internal and external factors.Methods:Thirty healthy adult SD rats were divided into normal control group,model group,and positive control group. A special carrot and spicy diet was used to rats and rats were occupied in the artificial climate box for a certain period of time. Body weights,food intake,anal temperature of all the rats were determined. Abdominal with drawal reflex and the parting grade of faeces shape were scored. The defecation frequency and stool water content were measured. The contents of TNF-α,IL-1β,DAO,D-lactate in serum,the SIgA content in colon mucosa and the MPO content in colon homogenates were assessed by radioimmunoassay. The number of Lactobacillus,Bifidobacterium,Enterococcus and Escherichia coli in the colon content of rats were measured and counted. The intestinal morphology was evaluated by histological examination. Results:After modeling,the rats body weight and food intake were decreased,and anal temperature was increased.The score of abdominal withdrawal reflex and the parting grade of faeces shape were increased,and the defecation frequency and stool water content were also increased. The contents of TNF-α,IL-1β,DAO,D-lactate in serum,the MPO content in colon homogenates were increased,but the SIgA content in colon mucosa was decreased. The numbers of Lactobacillus,Bifidobacterium were decreased. but the number of Enterococcus and Escherichia coli in the colon content of rats were increased. Compared with the normal group,the HPS score of the model group was significantly increased. After treatment with sulfasalazine(175 mg/kg) in 7 days,all the indexes were recovered.Conclusions:The stimulation of composite factors could be used to successfully generate the rat model of antibiotic related large intestine dampness-heat syndrome,its pathological characteristics and clinical symptoms were basically the same.
