摘要:目的:建立测定甘草中甘草苷、甘草酸及甘草多糖的方法,并比较不同采收时期甘草中甘草苷、甘草酸及甘草多糖的差异。方法:采用HPLC法测定甘草中甘草苷和甘草酸含量,色谱柱为Kromasil C_(18)柱(4.6 mm×250 mm,5μm);流动相为乙腈-0.05%磷酸溶液,梯度洗脱;体积流量为1 m L/min;检测波长为252 nm、276 nm;柱温为28℃;进样量为10μL,并采用紫外吸收光谱法测定甘草中甘草多糖。结果:甘草苷和甘草酸获得良好分离,在0.116 8-1.168 0μg及0.039 3-0.235 0μg与峰面积积分值呈良好线性关系,平均加样回收率分别为98.45%、98.34%,RSD分别为1.52%、1.78%,重现性试验RSD分别为0.35%、0.59%。甘草多糖分离良好,其重现性、稳定性亦良好,回收率较高。结论:本方法操作简便、结果准确、精密度好、分离度良好,为甘草药材的合理应用及其质量控制提供了依据。
Objective: To establish a determination method of liquirtin, glycyrrhizic acid and polysaccharide in licorice, and study the effect of different harvest period on their contents. Methods: HPLC method was applied for quality assessment of licorice with Kromasil C_(18) column(4.6 mm×250 mm, 5μm), flow-rate of 1.0 m L/min, column temperature of 28 ℃, mobile phase of acetonitrile and 0.05% phosphoric acid, respectively. The content of polysaccharide licorice was determined by ultraviolet absorption spectrometry. Results: The linear ranges of liquirtin and glycyrrhizic acid were 0.116 8-1.168 0 μg and 0.039 3-0.235 0 μg,respectively. The average recoveries of liquirtin and glycyrrhizic acid were 98.45%(RSD 1.52%) and 98.34%(RSD 1.78%).Licorice polysaccharide could be separated effectively with good repeatability, stability and recovery. Conclusion:Determination of these active components in licorice root with high specificity can be used to control its quality and assure lot to lot consistency.
