摘要:目的建立解酒保肝口服液的指纹图谱及多成分含量测定方法。方法采用高效液相色谱(HPLC)法和中药色谱指纹图谱相似度评价系统相似度软件建立11批解酒保肝口服液的指纹图谱,利用主成分分析(PCA)和偏最小二乘回归分析(PLS-DA)对解酒保肝口服液的质量进行分析研究;确定解酒保肝口服液中有效成分含量测定方法并测定其含量。结果建立了解酒保肝口服液HPLC指纹图谱,确定了29个共有峰,并指认检测了其中葛根素、毛蕊异黄酮苷、芦丁、木犀草苷、槲皮素5种有效成分。11批样品HPLC图谱相似度均大于0.950,PCA和PLS-DA找到了影响指纹图谱的6个主要成分,累计方差贡献率为92.89%,色谱峰24,29,22,23(木犀草苷),3,18,6,17,27,20,7,25,26,19(毛蕊异黄酮苷)的变量重要性投影(VIP)值>1,可作为解酒保肝口服液的质量差异性成分。11批解酒保肝口服液中葛根素、毛蕊异黄酮苷、芦丁、木犀草苷、槲皮素的质量浓度分别为106.698~236.795,0.815~1.868,14.552~16.662,8.204~13.12,18.731~20.427μg/mL。结论HPLC指纹图谱结合模式识别及多成分含量同时测定的方法为解酒保肝口服液提供了科学、可靠的质量评价方法,对解酒保肝口服液质量控制标准的建立具有重要意义,也为中药复方制剂的质量评价研究提供了参考。
Objective To establish a method for the determination of the fingerprint and multi-component content of Jiejiu Baogan Oral Liquid(解酒保肝口服液).Methods High-performance liquid chromatography(HPLC)and the similarity software of the Chinese medicine chromatographic fingerprint similarity evaluation system were employed to establish the fingerprint of 11 batches of Jiejiu Baogan Oral Liquid.Principal component ana-lysis(PCA)and partial least squares regression analysis(PLS-DA)were utilized to analyze the quality of Jiejiu Baogan Oral Liquid.The method for determining the content of the active components in Jiejiu Baogan Oral Liquid was determined and its content was measured.Results The HPLC fingerprint of the oral liquid was established,with 29 common peaks identified,and 5 active components,namely puerarin,pistil isoflavone glycoside,rutin,luteolin,and quercetin,were identified and detected.The similarity of the fingerprint spectra is greater than 0.95.PCA and PLS-DA identified six main components that affect the fingerprint spectra,with a cumulative variance contribution rate of 92.89%.The VIP values of chromatographic peaks 24,29,22,23(luteolin),3,18,6,17,27,20,7,25,26,and 19(pistil isoflavone glycosides)are greater than 1,which can be used as quality differential components of Jiejiu Baogan Oral Liquid.The mass concentrations of puerarin,pistil isoflavone glycoside,rutin,luteolin,and quercetin in 11 batches of Jijiu Baogan Oral Liquid were 106.698-236.795,0.815-1.868,14.552-16.662,8.204-13.12,and 18.731-20.427μg/mL,respectively.Conclusion The method of HPLC fingerprint combined with pattern recognition and simultaneous determination of multi-component content offers a scientific and reliable quality evaluation approach for Jiejiu Baogan Oral Liquid,which is of great significance for the establishment of quality control standards for Jiejiu Baogan Oral Liquid and also provides a reference for the quality evaluation of traditional Chinese medicine compound preparations.
