摘要:目的通过检测痛泻二草方对溃疡性结肠炎模型大鼠血清白细胞介素-2(IL-2)含量及肠黏膜一氧化氮(NO)含量、一氧化氮合酶(NOS)活性的影响,探讨痛泻二草方防治溃疡性结肠炎的作用机理。方法采用抗原免疫法和局部刺激法造成溃疡性结肠炎动物模型。将50只大鼠随机分为空白组、模型组、痛泻二草方组、痛泻要方组和补脾益肠丸组,分别对各组大鼠血清IL-2含量及肠黏膜NO含量、NOS活性进行检测。结果与空白组比较,模型组大鼠血清IL-2含量显著下降(P<0.01),肠黏膜NO含量、NOS活性显著升高(P<0.01,P<0.05);与模型组比较,痛泻二草方组可显著升高IL-2含量(P<0.05),降低结肠黏膜NO含量、NOS活性(P<0.01,P<0.05)。结论痛泻二草方具有调整结肠黏膜局部和机体免疫功能、调节结肠黏膜血流量、减少黏膜炎性细胞浸润、促进溃疡愈合的作用。
Objective To study effects of Tongxieercaofang (TXECF) on levels of IL-2 in serum and NO, NOS in intestinal mucosa tissue of rats with ulcerative colitis, and further to research the mechanism of restoreing ulcer of TXECF. Methods Ulcerative colitis model was established by immunization and stimulus method partly. 50 rats were divided into 5 groups randomly: normal group, model group, TXECF group, Tongxieyaofang (TXYF) group, Bupiyichangwan (BPYCW) group. Then the effect of TXECF on levels of IL-2 in serum and NO, NOS in intestinal mucosa tissue were observed. Results In successful model group, IL-2 in serum was much lower than that of normal group (P〈0.01), NO and NOS were much higher than those of normal group (P〈0.01, P〈0.05). Compared with model group, TXECF could not only increase IL-2 (P〈0.05), but also decrease NO and NOS in intestinal mucosa tissue significantly (P〈0.01, P〈0.05). TXECF has functions of adjusting immune function, regulating mucosal blood flow, easing inflammatory reaction and restoreing ulcer.
