摘要:目的探讨活血定眩胶囊对CSA大鼠血液ET、NO、6-Keto-PGF_(1α)、TXB_2及椎动脉NF-κB表达的影响。方法 SPF级大鼠72只,随机分为空白组、模型组、颈复康组、活血定眩胶囊低剂量组、中剂量组、高剂量组等6组,采用植骨压迫+组织离断联合造模。造模成功后连续灌胃8周,检测血液ET、NO、6-Keto-PGF_(1α)、TXB_2及椎动脉NF-κB表达。结果与空白组相比,血液中ET、TXB_2浓度增加,而NO、6-Keto-PGF_(1α)浓度降低,椎动脉中NF-κB表达增强,差异有统计学意义(P<0.05);与模型组相比,活血定眩胶囊高剂量组ET、TXB_2浓度降低,而NO、6-Keto-PGF_(1α)浓度增加,差异有统计学意义(P<0.01),椎动脉中NF-κB表达减弱,差异有统计学意义(P<0.05),活血定眩胶囊中剂量组及颈复康组ET、TXB_2浓度降低,而NO、6-Keto-PGF_(1α)浓度增加,差异有统计学意义(P<0.05),椎动脉中NF-κB表达减弱,但差异无统计学意义(P>0.05),活血定眩胶囊低剂量组各指标值均无统计学意义(P>0.05)。与颈复康组相比,活血定眩胶囊高剂量组各指标值差异有统计学意义(P<0.05),活血定眩胶囊中、低剂量组各指标值差异均无统计学意义(P>0.05)。结论活血定眩胶囊能有效改善椎动脉型颈椎病模型大鼠血液ET、NO、6-Keto-PGF_(1α)、TXB_2的表达,明显抑制NF-κB的表达。
Objective To explore the effects of Huoxuedingxuan capsule on expression of the NF-κB factor in arterial walls and ET,NO,6-Keto-PGF1αand TXB2 in arterial blood in CSA Rats. Methods 72 SPF Wistar rats were randomly divided into 6groups: blank group,model group,Jingfukang group and Huoxuedingxuan capsule groups( including low-dose group,middle-dose group and high-dose group). The CSA rat models were established by combining with bone graft compression and tissue amputation,the model of the CSA was successful,then start intragastric administration,for eight weeks. The concentrations of TXB2,NO,6-Keto-PGF1αand ET in arterial blood were detected; The expression of the NF-κB factor in arterial walls was detected. Results The model group compared with the blank group,the concentrations of ET and TXB2 increased in the blood,the concentrations of NO and 6-Keto-PGF1αdecreased in the blood,the expression of NF-κB increased in arterial walls,the differences were statistically significant( P〈 0. 05); The high-dose group compared with the model group,the concentrations of ET and TXB2 decreased,the concentrations of NO and 6-Keto-PGF1α increased,the differences were statistically significant( P〈 0. 01),the NF-κB expression decreased in the vertebral artery,the difference was statistically significance( P〈 0. 05),the middle-dose group and Jingfukang group compared with the model group,the concentrations of ET and TXB2 decreased,the concentrations of NO and 6-Keto-PGF_(1α) increased,the differences were statistically significant( P〈 0. 05),the expression of NF-κB decreased in the vertebral artery,but the difference was not statistically significant( P〉 0. 05); The low-dose group compared with the model group,each index had no statistical significance( P〉 0. 05). The high-dose group compared with the Jingfukang group,the differences in each index value were statistically significant( P〈 0. 05); The middle-dose group and low-dose group compared with the Jingfukang group,the differences in each index value were not statistically significant( P 〉0. 05).Conclusion Huoxuedingxuan capsule can effectively reduce the expression of NF-κB,ET and TXB2,promote the expression of NO and 6-Keto-PGF1α.
