摘要：胰岛素生长因子受体(IGF-1R)是一种酪氨酸激酶,在多种恶性细胞中高表达,主要通过介导凋亡保护、促进增殖和侵袭转移而加速肿瘤发展进程。在抗IGF-1R的研究中,分子靶向治疗虽提高了治疗效果,但耐药的产生仍然是治疗的主要挑战之一。中医药在抗耐药方面有独特的优势,近年来以IGF-1R为切入点,开展的大量中医药干预肿瘤的临床和实验研究取得了一定的成果。故本文就IGF-1R的分子结构及生物功能,对肿瘤的迁移、侵袭、增殖和凋亡调控作用及相关肿瘤信号传导,IGF-1R的靶向用药及耐药机制、中药单体和复方对IGF-1R的调控作用等方面进行归纳阐述,以期为肿瘤的临床治疗提供更多思路和参考。
IGF-1R,a tyrosine kinase,is highly expressed in a variety of malignant cells,which mainly accelerate the progression by promoting proliferation and invasion and inhibiting apoptosis of tumor cells.In the research of anti-IGF-1R,molecular targeted therapies have raised the effect of therapeutic,but inhibitor resistance is one of the major challenges.Traditional Chinese medicine(TCM)has unique advantages in drug resistance.In recent years,taking IGF-1R as the entry point,a large number of clinical and experimental studies on the intervention of TCM on the expression of IGF-1R in tumor cells have been carried out,and certain results have been achieved.Therefore,the molecular structure and biological function of IGF-1R,IGF-1R on tumor migration,invasion,proliferation and apoptosis regulation and related tumor signal transduction,IGF-1R targeted drug and drug resistance mechanism.At the same time,the regulatory effects of Chinese medicine monomer and Chinese medicine compound on IGF-1R were summarized and elaborated in order to provide more ideas and references for the clinical treatment of tumors.
