摘要:目的探讨归芪益元膏通过单磷酸腺苷激活的蛋白激酶/哺乳动物雷帕霉素靶蛋白(AMPK/mTOR)自噬信号通路对辐射旁效应损伤大鼠的保护作用及其机制。方法将32只SPF级SD大鼠随机分为正常对照组、单纯辐射组及归芪益元膏中、高剂量组,每组8只。归芪益元膏中、高剂量组预先灌胃归芪益元膏[3.28g/(kg·d)、6.56g/(kg·d)],其余各组予等量0.9%氯化钠溶液灌胃,连续干预14d。干预14d后,单纯辐射组及归芪益元膏中、高剂量组大鼠右肺予8Gy剂量X射线单次照射,铅皮屏蔽其余身体部位;正常对照组不给予照射。照射后处死所有大鼠,免疫组化、qPCR检测大鼠右肺、左肾组织中的p62、Beclin1、AMPK、mTOR蛋白及mRNA表达情况。结果单纯辐射组右肺、左肾组织中的p62蛋白及mRNA表达量明显低于正常对照组,Beclin1、AMPK、mTOR蛋白及mRNA表达量明显高于正常对照组(P<0.01)。归芪益元膏中、高剂量组右肺、左肾组织中的p62蛋白及mRNA表达量明显高于单纯辐射组,Beclin1、AMPK、mTOR蛋白及mRNA表达量明显低于单纯辐射组(P<0.05)。结论辐射旁效应损伤大鼠肺、肾组织中自噬通路AMPK/mTOR,造成自噬蛋白p62、Beclin1表达异常。归芪益元膏可以调控AMPK/mTOR信号通路及自噬蛋白p62、Beclin1表达,抑制自噬,从而发挥对辐射诱发体内旁效应损伤的防护作用。
Objective To investigate the protective effect and mechanism of Guiqi Yiyuan ointment on rats injured by radiation induced bystander effect through AMP activated protein kinase/mammalian target of rapamycin(AMPK/mTOR)autophagy signaling pathway.Methods Thirty-two SPF SD rats were randomly divided into normal control group,simple radiation group,Guiqi Yiyuan ointment medium and high dose groups,with 8 rats in each group.The Guiqi Yiyuan ointment medium and high dose groups were pre-administered with Guiqi Yiyuan ointment[3.28g/(kg·d),6.56 g/(kg·d)],and the other groups were given the same amount of 0.9%sodium chloride solution for 14 d.After 14 d of intervention,the right lung of rats in the simple radiation group,Guiqi Yiyuan ointment medium and high dose groups were irradiated with single dose of 8 Gy X-ray,and the rest of the body parts were shielded by lead skin;the normal control group was not irradiated.All rats were sacrificed after irradiation.The expression of p62,Beclin1,AMPK,mTOR protein and mRNA in the right lung and left kidney of rats were detected by immunohistochemistry and qPCR.Results The expression of p62 protein and mRNA in the right lung and left kidney tissues of the simple radiation group were significantly lower than those of the normal control group,and the expression of Beclin1,AMPK,mTOR proteins and mRNA were significantly higher than those of the normal control group(P<0.01).The expression of p62 protein and mRNA in the right lung and left kidney of the Guiqi Yiyuan ointment medium and high dose groups were significantly higher than those of the simple radiation group,and the expression of Beclin1,AMPK and mTOR proteins and mRNA were significantly lower than those of the simple radiation group(P<0.05).Conclusion The autophagy pathway AMPK/mTOR in lung and kidney tissues of rats was damaged by radiation induced bystander effect,resulting in abnormal expression of autophagy protein p62 and Beclin1.Guiqi Yiyuan ointment can regulate AMPK/mTOR signaling pathway and autophagy protein p62 and Beclin1 expression,inhibit autophagy,and thus plays a protective role in damage by radiation induced bystander effect.
