摘要:目的:探讨敦煌医方泻肺汤治疗特发性肺纤维化的作用机制。方法:采用Szapiel法复制大鼠肺纤维化模型以泻肺汤进行干预;肺功能测定仪检测吸气时间(inspiratory time,TI)、呼气时间(expiratory time,TE)、潮气量(tidal volume,TV)、肺泡通气量(alveolar ventilation,AV),ELISA法检测转化生长因子-β_1(transforming growth factor-β_1,TGF-β_1)和肿瘤坏死因子-α(tumour necrosis factor-α,TNF-α)含量,比色分析法测定一氧化氮合成酶(nitric oxide synthase,NOS)活性和一氧化氮(nitric oxide,NO)含量,RT-PCR法和Western-blot法测定bcl-2/bax基因表达及Bcl-2/Bax蛋白表达。结果:与正常对照组比较,模型组大鼠TE、TV和AV等指标下降, bcl-2基因和蛋白表达均降低,TNF-α含量、TGF-β_1含量、NOS活性、NO含量、bax基因和蛋白表达均升高。与模型组比较,泻肺汤各干预组大鼠TV和AV升高,高剂量组大鼠TE显著升高;泻肺汤中、高剂量组bcl-2基因和蛋白表达均升高,TNF-α含量、TGF-β_1含量、NOS活性、NO含量、bax基因和蛋白表达均降低。结论:泻肺汤通过下调Bax和上调Bcl-2表达调控氧化应激反应,从而发挥抗肺纤维化作用。
Objective: To explore the mechanism of Xie Fei decoction in the treatment of idiopathic pulmonary fibrosis(IPF). Methods: The IPF rats were caused by Bleomycin and treated by Xie Fei decoction. The lung function indexes of TI,TE,TV and AV were checked by function testing instrument, the contents of transforming growth factor(TGF-β1) and tumour necrosis factor(TNF-α) in serum of rats were detected by enzyme linked immunosorbent assay(ELISA), the activity of nitric oxide synthase(NOS) and the contents of nitric oxide(NO) in bronchoalveolar lavage fliud(BALF) of rats were detected by colorimetric assay, the gene expression of bcl-2 and bax in lung of IPF rats was detected by real-time PCR, the protein expression of Bcl-2 and Bax were detected by Western-blot. Results: Compared with the namor contral group, the lung function indexes of TE、TV and AV were decreased obviously in model group;The contents of TNF-α and TGF-β1 in serum were increased significantly in model group, the activities of NOS and the contents of NO in BALF were also raised obviously in model group, the gene expression of bax and the protein expression of Bax in lung tissue were increased obviously in model group, yet the gene expression of bcl-2 and the protein expression of Bcl-2 in lung tissue were decreased obviously in model group. Compared with the model group, the lung function indexes of TV and AV were decreased obviously in Xie Fei decoction group, the lung function index of TE was decreased obviously in high-dose group of Xie Fei decoction;The contents of TNF-α and TGF-β1 in serum were decreased significantly in middle-dose,high-dose group of Xie Fei decoction, meantime, the activities of NOS and the contents of NO in BALF were also dropped obviously in middle-dose,high-dose group of Xie Fei decoction, as well as, the gene expression of bax and the protein expression of Bax in lung tissue were declined obviously in middle-dose,high-dose group of Xie Fei decoction, but the gene expression of bcl-2 and the protein expression of Bcl-2 in lung tissue were increased obviously in middle-dose,high-dose group of Xie Fei decoction. Conclusion: Xie Fei decoction could regulate oxidative stress response by down regulation of Bax and up regulation of Bcl-2 and plays a protective effect on pulmonary fibrosis.
