摘要:目的:对湿生扁蕾乙醇提取物进行血清药物化学研究,探讨湿生扁蕾乙醇提取物的药效物质基础。方法:通过高效液相色谱法(HPLC)在相同的色谱条件下分别测定湿生扁蕾乙醇提取物、空白血清和不同灌胃剂量的给药血清的HPLC图谱,通过比较各成分的相对保留时间,确定湿生扁蕾乙醇提取物大鼠灌胃后血中移行成分及不同给药剂量湿生扁蕾含药血清中血中移行成分的差异。结果:随着给药剂量的增加,湿生扁蕾含药血清中的血中移行成分及其含量也相应增加。在湿生扁蕾给药剂量为5.16g/kg时,给药血清中发现7种移行成分,其中4种为原型成分,且检出3种分别为当药黄素,当药醇苷和木犀草素,另外3种可能为原型成分的次生代谢产物或新生代谢产物。结论:给药剂量会对湿生扁蕾的血中移行成分的种类和含量有较大影响,7种血中移行成分可能为湿生扁蕾的药效成分,但是有些成分含量比较低,是否能成为有效成分有待继续考究,对其进行进一步研究有助于阐明湿生扁蕾的药效物质基础和作用机理。
Objective: By studying the serum pharmacochemistry of ethanol extract of Gentianopsis paludosama,study on material base of the effect of ethanol extract of Gentianopsis paludosama. Methods: HPLC chromatography of ethanol extract of Gentianopsis paludosama,blank serum and different intragastric doses of serum containing drug under the same chromatographic conditions were determined. Components whose peaks shifted and differences of which from rats who were given the different doses of Gentianopsis paludosama were determined. Results:With the increase of dosages,components in the serum and contents of which also increased. 7 components were found in the serum of ethanol extract of Gentianopsis paludosama,4 of which were prototype components,including swertisin,swertianolin and luteolin,and the other 3 were maybe secondary metabolites or new metabolites of prototype components. Conclusion: The dose has the great influence on types and contents of components in the blood of Gentianopsis paludosama and 7 components may be effective components of Gentianopsis paludosama. However,some components of them are very low. Further studies should be used to determine whether or not they are active ingredients.
