摘要:目的探讨C-型利钠肽(CNP)对大鼠液体胃排空和小肠传输的影响。方法 70只健康雄性Wistar大鼠随机分为治疗组(42只)和对照组(28只),取治疗组37只和对照组23只分别经腹腔注射CNP和生理盐水30 min后用葡萄糖酚红溶液灌胃,灌胃后10、20、30 min分批处死大鼠,切取胃和小肠近、中、远各段留取内容物后使用分光光度计测定胃肠各段酚红残留率。另取治疗组和对照组大鼠各5只在灌胃前2 h分别腹腔注射奥美拉唑,灌胃后10min处死大鼠测定胃内酚红残留率。结果灌胃后10、20、30 min治疗组胃内酚红残留率分别为66.1%±1.7%、46.5%±1.8%和38.2%±3.1%,对照组为50.2%±2.0%、29.8%±1.1%和21.7%±1.3%,治疗组胃内酚红残留率均多于对照组(P<0.05);比较治疗组和对照组小肠近段酚红残留率差异无统计学意义;灌胃后10、20min治疗组中段小肠酚红残留率分别为1.2%±3.4%和18.7%±3.1%,对照组分别为11.2%±1.6%和33.7%±1.5%,治疗组酚红残留率少于对照组(P<0.05);灌胃后10、20、30 min治疗组远段小肠酚红残留率分别为0.2%±0.9%、1.9%±0.1%和2.7%±2.4%,对照组分别为2.7%±0.6%、5.9%±1.4%和7.8%±2.1%,两组比较治疗组酚红残留率低于对照组(P<0.05);奥美拉唑预处理灌胃后10 min治疗组较对照组胃内酚红残留率增多,分别为62.1%±4.7%和49.8%±6.4%(P<0.05),与未给予奥美拉唑预处理组比较,差异无统计学意义。结论 CNP可使大鼠液体胃排空减慢,小肠传输延迟。
Objective To assess the effects of C-type natriuretic peptide(CNP) on gastric emptying and gastrointestinal transit of liquid in awake rats.Methods The male Wistar rats were randomly divided into CNP treatment group and control group.The rats in treatment group received injection of CNP(1 mg/kg),while the rats of control group only received an equal volume saline.After 24 h fast,they were gavage fed(1.5 mL) with phenol red in 5%glucose solution and then sacrificed respectively in 10,20,30 min later.Dye recovery in the stomach,proximal,mid,and distal small intestine was obtained by spectrophotometry.To rule out the effect of gastric secretion changes,other group was omeprazole-pretreated before gavage fed.Results Gastric dye recovery of the CNP treatment group was greater than that of the control(66.1%±1.7%vs50.2%±2.0%,46.5%±1.8%vs 29.8%±1.1%and 38.2%±3.1%vs 21.7%±1.3%) in all subsets,respectively,at 10,20,30 min(P <0.05).There were no differences in proximal intestine retention between CNP treatment groups and control groups in all subsets,at 10,20,30 min.In mid small intestine dye recovery of the CNP treatment group was decreased compared to the control(1.2%±3.4%vs 11.2%±1.6%and 18.7%±3.1%vs 33.7%±1.5%) in all subsets,respectively,at 10,20 min(P <0.05).In distal small intestine dye recovery of the CNP treatment group was decreased compared to the control(0.2%±0.9%vs 2.7%±0.6%,1.9%±0.1%vs 5.9%±1.4%and2.7%±2.4%vs 7.8%±2.1%) in all subsets,respectively,at 10,20,30 min(P <0.05).Omeprazolepretreated did not alter this phenomenon.Gastric dye recovery of the CNP treatment group was greater than that of the control(62.1%±4.7%vs 49.8%±6.4%) at 10 min(P <0.05).Conclusion The CNP delays the gastric emptying and gastrointestinal transit of liquid in awake rats.
