摘要:目的:观察甘肃产藏药五脉绿绒蒿有效部位对肝纤维化大鼠TGF-β1表达的影响。方法:在给药的基础上,采用sc CCl4、喂饲高脂低蛋白复合饲料并饮用20%乙醇来复制肝纤维化动物模型,6周后对大鼠肝组织免疫组化染色,检测TGF-β1的表达,同时检测血清中AST、ALT和TGF-β1的水平以及肝组织中Hyp的含量。结果:复合因素肝纤维化造模结束后,与模型对照组比较,甘肃产藏药五脉绿绒蒿醇提物、黄碱组以及总黄酮能明显降低血清ALT、AST以及肝组织中Hyp与胶原蛋白的含量,显著降低血清与肝组织中TGF-β1的表达,同时发现总黄酮的作用有一定的量效关系。结论:甘肃产藏药五脉绿绒蒿醇提物及其中的总黄酮对CCl4复合因素诱导的肝纤维化大鼠具较好的保护作用,对TGF-β1表达的抑制作用可能是其作用机制之一。
Objective:To investigate the influence of active fractions of Meconopsis quintuplinervia Regel (MQR) growing in Gansu Province on TGF-β1 of experimental liver fibrosis in rats. Methods:Under drug's effects, using sc CC14, eating high-fat low- protein diet and drinking 20% alcohol in six weeks, the model of hepatic fibrosis rats was established to valuate the effects of the ex- tractive parts from MQR through detecting indexes in serum and hepatic tissue and expression of TGF-β1. Results:The extractive of ethanol, total flavones and mixture of total flavones and total alkaloids from MQR growing in Gansu Province could significantly de- crease ALT, AST and TGF-βI in serum and Hyp and collagen protein in hepatic tissue, and they could also inhibit the expression of TGF-βI in the liver tissue of the rats. The effects of total flavones exhibit the dose-effect relationship. Conclusion:The ethanol extrac- tive from MQR growing in Gausu Province and total flavones in it have better protecting effects on hepatic fibrosis in rats induced by carbon tetrachloride compounded other factors, and the one of mechanisms maybe that it can inhibit the expression of TGF-~I in hepat- ic fibrosis tissue.
