摘要:目的探讨地龙蛋白对糖尿病性勃起功能障碍(DMED)大鼠的干预作用及其机制。方法60只大鼠随机选取10只作为空白组,其余50只通过腹腔注射链脲佐菌素(STZ)构建糖尿病大鼠模型,8周后使用阿朴吗啡(APO)诱导筛选符合DMED症状的大鼠模型,将DMED大鼠随机分为模型组、西地拉非组及地龙蛋白低、中、高剂量组,相应药物干预4周后,检测大鼠平均颈动脉压(MAP)和阴茎海绵体最大内压(ICPmax)以评估大鼠勃起功能,试剂盒检测阴茎海绵体组织SOD活性及NO、cGMP、MDA水平,RT-qPCR法检测阴茎海绵体组织eNOS、cGMP mRNA表达,免疫组化和Western blot法检测eNOS蛋白表达,HE染色观察阴茎海绵体病理结构的改变。结果与空白组比较,模型组大鼠血糖、阴茎海绵体组织MDA水平升高(P<0.01),体质量、ICPmax/MAP、阴茎海绵体组织SOD活性及NO、cGMP水平、阴茎海绵体组织eNOS mRNA和蛋白表达均降低(P<0.01);与模型组比较,各给药组以上指标均有改善(P<0.01)。空白组阴茎海绵体组织结构正常;模型组阴茎海绵体组织肌纤维、胶原纤维断裂,拧结明显;各给药组阴茎海绵体组织病理损伤较模型组均有不同程度的改善。结论地龙蛋白具有改善糖尿病大鼠勃起功能障碍的作用,其机制可能与抑制氧化应激、上调大鼠阴茎海绵体NO-cGMP通路表达有关。
AIM To investigate the interventive effects of earthworm protein on diabetic erectile dysfunction(DMED)in rats and the mechanism.METHODS Among the the sixty rats,ten were randomly divided into the control group,and the other 50 rats were induced into diabetic rat models by intraperitoneal injection of streptozotocin(STZ),followed by eight weeks later apomorphine(APO)induction and the examination of the rats conformity to the symptoms of DMED.The DMED rats were randomly divided into the model group,the siderafil group,and the earthworm protein groups for low dose,medium dose,and high dose interventions.After four weeks of corresponding drug use,the rats had their erectile function assessed by measurement of mean carotid pressure(MAP)and the maximum internal pressure of the corpus cavernosum(ICPmax);their SOD activity and levels of NO,cGMP and MDA in the corpus cavernosum tissue detected by kits;their mRNA expressions of eNOS and CGMP in the corpus cavernosum tissue detected by RT-qPCR;their eNOS protein expression detected by immunohistochemistry and Western blot;and their changes of pathological structure of the corpus cavernosum observed by HE staining.RESULTS Compared with the control group,the model group displayed in the corpus cavernosum increased blood sugar and MDA level(P<0.01),and decreased body weight,ICPmax/MAP,SOD activity and NO,cGMP level,eNOS mRNA and protein expression(P<0.01).Compared with the model group,each drug group found improvement in terms of the aforementioned indexes levels(P<0.01).The tissue structure of the cavernous body was found to be normal in the control group;to be with broken and obviously kinked muscle fibers and collagen fibers in the model group;to be with pathological penile tissue damage improved to various degrees in the treatment groups.CONCLUSION Earthworm protein can improve erectile dysfunction in diabetic rats,and its mechanism may be related to the inhibition of oxidative stress and up-regulation of NO-cGMP signal pathway expression in the rat cavernous body.
