摘要:目的:比较甘肃产柴胡不同炮制品中柴胡皂苷a的含量及其对小鼠的抗抑郁作用。方法:采用HPLC测定甘肃产柴胡不同炮制品中柴胡皂苷a含量,并通过自主活动、悬尾和强迫游泳实验及脑内5-HT及NE含量的测定,比较柴胡不同炮制品对行为绝望抑郁模型小鼠的影响。结果:甘肃产柴胡不同炮制品中柴胡皂苷a的含量以酒柴胡含量最高,鳖血柴胡次之。不同炮制品均能缩短两种行为绝望模型小鼠的不动时间,以鳖血柴胡及醋柴胡效果最明显;不同炮制品均可提高小鼠脑内5-HT含量,以鳖血柴胡效果最显著,具有显著性差异。结论:炮制可使甘肃产柴胡皂苷a含量发生变化,并改善小鼠的绝望行为,增加脑内5-HT含量而显示抗抑郁作用,有良好的开发前景。
Objective: To determine saikosaponin-a in different processed products of Radix bupleuri in Gansu Province and study its antidepressant effect. Methods: HPLC was used to test the content of saikosaponin-a in different processed products of Radix bupleuri and the antidepressant effects were observed by using tile spontaneous activities test, tail suspension test. forced swimming test in mice, and 5-HT and NE in brain were detected and to be compared. Results: The content of saikosaponin-a in wine processing Radix bupleuri was the highest, then was soli-shelled turtle blood-baked ones. Different processed products could all reduce the immobility time of mice in two kinds of behavioral despair model, especially soft-shelled turtle bloodbaked ones and vinegar-baked ones. Different processed products could all increase the 5-HT level in mice brain, especially soft-shelled turtle blood-baked ones. Conclusion: Radix bupleuri in Gansu Province has promising developmenl prospect. Processing can change its saikosaponin-a content and enhance its antidepressant effect by improving behavioral despair of mice and increasing the 5-HT level in brain.
