摘要:目的:研究甘肃产素花党参水煎剂抗皮肤衰老的作用和机制。方法:昆明种小鼠随机分为模型组、空白对照组、党参低剂量组和党参高剂量组、阳性对照组,经颈背部皮下注射5%D-半乳糖连续6 w,建立小鼠亚急性衰老模型,在光镜及电镜下分别观察党参水煎剂对D-半乳糖致衰老小鼠皮肤组织形态的影响;检测小鼠皮肤组织丙二醛(MDA)含量、脂褐素(LF)含量、超氧化物歧化酶(SOD)活性、ATP酶活性及羟脯氨酸(HYP)的含量。结果:党参低剂量组(5g/kg)及党参高剂量组(15g/kg)均可改善皮肤组织形态,光镜下,党参低剂量组、高剂量组皮肤真皮层显著增厚,纤维成分增多,排列较紧密,皮肤形态学结构较接近空白对照组;电镜下,党参低剂量组皮肤组织细胞可见轻度损伤,细胞器肿胀及少量空泡形成,党参高剂量组皮肤组织细胞结构完整,接近空白对照组皮肤组织细胞;MDA含量和LF含量降低,SOD活性及ATP活性升高,HYP含量升高。结论:甘肃产素花党参水煎剂能改善皮肤组织病理形态和自由基代谢失调的状况,对皮肤有一定的抗衰老作用。
Objective: To investigate the mechanism of Codonopsis pilosula var. modesta decoction on the skin. Method: SPF mice were evenly randomize into the blank control group,the model group,positive control group( the model + VE group),the model + Codonopsis pilosula var.modesta of high dose group( 5g /kg) and the model + Codonopsis pilosula of low dose group( 15g /kg),sub-acutely aging model was estbalished by injecting of 5% D-galactose into neck subcutaneously for consecutive 6w. Observe morphological changes of aging mice induced by D-galactose. Detect the level of malondialdehyde( MDA),lipofuscin( LF),superoxide dismutase( SOD),ATP enzyme and hydroxyproline( HYP) of entaneou tissue. Result: Under microscope,in the model + low dose Codonopsis pilosula var. modesta group and the model + high dose Codonopsis pilosula var. modesta group,the dermis layer of skin was thicker,fiber increased significantly with close arrangement,and skin morphological structure was closer to the normal control group. MDA and LF were lower significantly( P < 0. 05). SOD and ATP activity increased considerably( P < 0. 05). HYP rose( P < 0. 05). Conclusion: Gansu Codonopsis pilosula var. modesta have anti-aging effect in improving the pathological change and balancing free radical metabolism,and the high dose effect was better than that of low dose.
