摘要:目的观察去卵巢大鼠骨Ⅰ型胶原表达变化及温和灸、苯甲酸雌二醇注射液(BE2)的干预效果,探讨"补虚化瘀"灸法治疗绝经后骨质疏松症(PMO)的机制。方法将50只8月龄雌性Wistar大鼠随机分为假手术组10只和去势组40只,后者行双侧卵巢切除术,并于90 d后随机分为模型组、雌激素组、艾灸组和艾灸+雌激素组各10只,其后假手术组和模型组正常饲养,雌激素组予BE2后肢肌肉注射、艾灸组按"补虚化瘀法"取穴行温和灸治疗,艾灸+雌激素组同时给予BE2注射和艾灸治疗。疗程结合后处死大鼠,分别以免疫组化法和原位杂交法检测右股骨Ⅰ型胶原mRNA表达。结果模型组大鼠右股骨Ⅰ型胶原蛋白及mRNA表达均明显低于假手术组及各治疗组,各治疗组中仅艾灸+雌激素组mRNA表达阳性反应面积明显大于艾灸组(P<0.05或0.01),余各指标比较无显著差异。结论温和灸可明显增强去卵巢大鼠股骨Ⅰ型胶原表达(效果与BE2相似),此可能为"补虚化瘀"灸法治疗PMO的机制之一。
Objective To observe the expression changes of collagen type I in ovariectomized rat femur model and the intervention effects of mild moxibustion and estradiol benzoate injection ( BE= ), and to investigate possible mechanism of tonifying deficiency and dissolving blood stasis in treating postmenopausal osteoporosis (PMO). Methods Of 50 female Wistar rats at 8-month old, 10 were randomly allocated to the sham group and other 40 underwent the ovariectomized opera- tion. After 90 days, the ovariectomized female Wistar rats were randomly divided into 4 group, 10 in each group : ovx group (model group), estrogen group, moxibustion group and moxibustion estrogen group. The rats in the sham group and ovx group did not received any treatment. The rats in the estrogen group received the treatment of muscle injection of estradiol benzoate in the leg. The rats in moxibustion group received the treatment of mild moxibustion according to the method of " tonifying deficiency and dissolving blood stasis" to select points. The rats in moxibustion estrogen group received the treat- ment of the mild moxibustion and muscle injection of estradiol benzoate. The rats were killed at the end of course to observe bone collagen type I and bone collagen type I gene mRNA expression by using immunohistochemistry and in situ hybridiza- tion. Results The expression of collagen type I and collagen type I mRNA in the model group was significantly lower than that of the sham group and other treatment groups; during the treatment groups, the positive reaction area in the moxi- bustion estrogen group was significantly larger than that of the moxibustion group ( P 〈 0.05 or P 〈 0.01 ), and the other in- dexes had no difference. Conclusion The mild moxibustion can improve the expression level of collagen type I and collagen type I mRNA of the ovariectomized rats, and this may be one of the mechanisms of mild moxibustion by tonifying defi- ciency and dissolving blood stasis in treatment of PMO.
