摘要:[目的]观察芍药苷治疗溃疡性结肠炎(ulcerative colitis,UC)的疗效,并探讨其作用机制。[方法]40只大鼠中10只为空白组,30只采用TNBS/乙醇灌肠法制备UC大鼠模型后分为模型组、芍药苷组及SASP组,光镜观察各组结肠组织病理形态变化并评分,采用ELISA法检测各组大鼠血清白细胞介素(IL)-2、IL-10、IL-6水平。[结果]与空白组大鼠比较,模型组大鼠血清IL-2[(43.05±8.42)ng/L]、IL-6[(248.05±46.56)ng/L]含量均明显升高,IL-10[(23.47±7.66)ng/L]含量显著下降,差异具有统计学意义(P<0.01);芍药苷组血清IL-2[(28.25±1.25)ng/L]、IL-6[(186.56±47.14)ng/L]水平均较模型组降低,而IL-10[(42.29±5.11)ng/L]水平明显升高,差异具有统计学意义(P<0.05、P<0.01)。[结论]芍药苷能够通过下调促炎因子IL-2、IL-6的表达,升高抗炎因子IL-10的表达,促进抗炎和抑炎因子的平衡,调节肠道异常免疫反应、抑制炎症反应,从而修复溃疡。
[Objective]To observe the effect of paeoniflorin on ulcerative colitis (UC) and to investigate the possible mechanism. [Methods]The UC rat models were prepared by the TNBS/ethanol enema, then randomly divided into three groups of model group,paeoniflorin group and SASP group (each n= 10) and 10 rats as normal control. The colon tissue pathological morphology was observed and scored under the light microscope,and the levels of IL-2, IL-10 and IL-6 were detected in rat serum. [Results]Compared with the control group, the content of IL-2 ([43.05 ± 8. 42] ng/L), IL-6 ([248.05± 46.56] ng/L) was markedly higher, and that of IL-10([23.47±7.66] ng/L)was decreased significantly in rats of the model group (P〈0. 01). The content of IL-2([28. 25-±t-1.25] ng/L)and IL-6([186.56_±47.14] ng/L) in rats of the paeoniflorin groups were lower, and that of IL-10(42.29±5. 111 ng/L) was higher than those of the model group respectively (P〈0.05, P〈0.01). [Conelusion]The paeoniflorin can decrease the expression of IL-2, IL-6,and increase the expression of IL-10, which might be one of the mechanisms of inhibition of inflammation for UC.
