摘要:目的:探讨肺心通对慢性肺源性心脏病(CPHD)大鼠的治疗效果及其作用机理。方法:50只Wistar大鼠,雌雄各半,随机分为空白组、模型组、肺心通高、中、低剂量组,以2%野百合碱(MCT)按60 mg/kg 1次腹腔注射复制CPHD大鼠模型。同时给模型组、肺心通高、中、低剂量组灌胃相应药物,连续用药21 d。测定大鼠血清和肺组织匀浆中一氧化氮(NO)、内皮素-1(ET-1)的含量;测定肺小动脉及支气管平滑肌血管内皮细胞生长因子(VEGF)的表达;观察肺组织病理形态学改变。结果:肺心通可显著提高CPHD模型大鼠的NO含量;降低ET-1的含量;降低大鼠肺小动脉及支气管平滑肌中VEGF的表达;改善肺组织的病理改变。结论:肺心通对CPHD大鼠有较好的治疗作用。
Objective:To research the functionary mechanism and treating effect of Feixintong(FXT) on chronic pulmonary heart disease( CPHD). Methods:50 Wister rats were randomly separated into five groups:blank control group, model group, FXT groups of high, middle and low dosage, The rat model of corpulmonale was established by injecting 2 % monocrotaline(60 mg/kg)i, p. At the same time, model group, FXT groups of high, middle and low dosage were separately administrated with physical saline or FXT for 21 days. NO, ET-1 and expression of VEGF in bronchi arterioles was measured. Lung tissue sections were examined to study the morphological alternations. Results: FXT could evidently improve the NO content of rat model of CPHD; reduce the ET-1 content: reduce the expression of VEGF in bronchi arterioles. Conclusions:FXT treatment is effective on rat model of CPHD.
