摘要:Objective To observe the effects of catgut-embedding on depressive-like behaviors, synthesis and transport of 5-hydroxytryptamine (5-HT) neurotransmitters, and expression levels of 5-hydroxytryptamine 1A receptor (5-HT1AR) in the dorsal raphe nucleus (DRN) and hippocampus of depressed rats, and to explore the mechanism of point catgut-embedding on depression. Methods Ten rats were selected from 60 male Sprague-Dawley rats as a blank group using the random number table method, and the remaining 50 rats were prepared to be a depression model of "isolation rearing combined with chronic unpredictable mild stimulation". Ten rats that failed to establish the model were excluded, and the remaining rats that successfully established the model were randomly divided into a model group, a grasp group, a fluoxetine group, and a catgut-embedding group, with 10 rats in each group. The grasp group was given the same grasp as the intervention groups. The fluoxetine group was given fluoxetine solution [2 mg/(kg & centerdot;bw)] by gavage once daily for 3 weeks. The catgut-embedding group was given catgut-embedding at points Baihui (GV20), Shenmen (HT7), Neiguan (PC6), and Taichong (LR3) for "soothing the liver and regulating the mind" once a week for 3 consecutive weeks. The behavioral changes of rats were observed by forced swimming and open field test (OFT). The morphological changes of DRN and hippocampus were observed by hematoxylin-eosin staining. The levels of DRN and hippocampal 5-HT transmitters were detected by enzyme-linked immunosorbent assay. Western blotting was used to detect the expression levels of tryptophan hydroxylase (TPH) 2, serotonin transporter (SERT), and 5-HT1AR in the DRN and hippocampus. Results Compared with the blank group, the total distance, average speed, and high activity percentage of OFT were significantly reduced (P<0.05); the low activity percentage of forced swimming was increased significantly (P<0.05) in the model group; the numbers of DRN and hippocampal neurons were decreased, and nucleus was deeply stained and pyknotic; the levels of DRN, 5-HT, and TPH2 were significantly decreased (P<0.05), and SERT protein expression was significantly increased (P<0.05) in the hippocampus; 5-HT1AR protein expression in the DRN was significantly increased (P<0.05), and 5-HT1AR protein expression in the hippocampus was significantly decreased (P<0.05) in the model group. The cytoplasm showed hyaline degeneration with disappeared nuclei and lysed Nissl bodies in DRN and hippocampal neurons in the grasp group. Compared with the grasp group, the total distance, average speed, and high activity percentage of OFT were significantly increased (P<0.05) in the fluoxetine and catgut-embedding groups, while the low activity percentage of forced swimming was significantly decreased (P<0.05); the numbers of DRN and hippocampal neurons were decreased slightly, and the nucleus was slightly irregular; the expression levels of DRN and hippocampal TPH2 protein were significantly increased (P<0.05), and the expression level of SERT protein was significantly decreased (P<0.05); the expression level of 5-HT1AR protein in the DRN was significantly decreased (P<0.05), and the expression level of 5-HT1AR protein in the hippocampus was significantly increased (P<0.05) in rats in the fluoxetine group and the catgut-embedding group. Conclusion Catgut-embedding based on "soothing the liver and regulating the mind" can effectively improve depressive-like behaviors of depressed model rats, repair the injury of DRN and hippocampal neurons, improve and regulate the synthesis, transport, and key receptor expression of DRN-hippocampal 5-HT transmitters. Its mechanism may be related to the regulation of the serotonergic system.
