摘要:目的观察慢性阻塞性肺疾病大鼠模型病变发展的情况。方法将40只SPF级Wistar大鼠随机分为空白组、模型1组、模型2组、模型3组,每组10只,运用气管内滴注脂多糖加烟熏法建立慢性阻塞性肺疾病大鼠模型。观察或检测各组大鼠一般状态及体质量、呼吸功能、肺组织形态的变化。结果与空白组比较,各模型组大鼠精神状态差,食量及活动减少,体质量增幅减小,肺干、湿质量比显著降低(P<0.01),肺部病理损害明显,呼吸功能指标改变显著(P<0.01);模型2组与模型3组比较,大鼠一般状态、体质量、呼吸功能及肺组织形态等指标比较差异均无统计意义(P>0.05)。结论模型1组病变呈慢性支气管炎合并呼吸气流受限,模型2组、3组病变呈慢性支气管炎或(和)肺气肿合并呼吸气流受限;用2次气管内滴注脂多糖加烟熏28 d方法所建立的慢性阻塞性肺疾病大鼠模型可靠,造模停止2周后模型稳定。
Objective To observe the pathological changes in the rat model with chronic obstructive pulmonary disease( COPD). Methods 40 Wistar rats( SPF) were randomly divided into four groups: the normal group,the model group 1( 14 days),the model group 2( 28 days) and the model group 3( 42 days),10 rats in each group. COPD rat model was induced by cigarette smoking inhalation plus intratracheal LPS exposure. General status,body weight,lung pathological lesion and respiratory function of all rats were observed and detected. Results Compared with the normal group,rats in other model groups were short of mental state. Their appetite,activities fell off and the body weight increased slowly. D / W,MAN and RF decreased significantly( P < 0. 01); the lung tissue impaired obviously. There was no significant difference between model group 2 and model group 3. Conclusion Pathological changes in model group 1 appear chronic bronchitis combined with limitation of respiratory air current,the inhalation is light; pathological changes in group 2 and group 3 appear CB or emphysema combined with limitation of respiratory air current,the inhalation is heavy. COPD rat model that was induced by cigarette smoking inhalation and intratracheal LPS exposure is reliable. The model is stable when the impairmental factors were eliminated after two weeks.
