摘要:目的探究红芪总多糖对脑缺血再灌注损伤大鼠海马组织的保护作用。方法按照体重将Wistar大鼠随机分为6组:假手术组、模型组、对照组和低、中、高3个剂量实验组,每组25只。造模前,低、中、高3个剂量实验组每日分别灌服红芪总多糖(50,100,200 mg·kg^-1),对照组每日灌服尼莫地平15mg·kg^-1,假手术组和模型组灌胃给予等体积0.9%Na Cl,均连续10 d。用线栓法建立大鼠脑缺血再灌注模型。以酶联免疫吸附实验测定脑组织的白细胞介素-10(IL-10)、IL-6和肿瘤坏死因子-α(TNF-α)水平。结果假手术组、模型组、对照组和低、中、高3个剂量实验组的TNF-α含量分别为(35.46±4.31),(58.57±6.23),(39.88±5.46),(58.38±5.87),(56.43±6.27)和(48.19±5.35)ng·L^-1;这6组的IL-10含量分别为(35.24±4.22),(46.43±4.59),(38.72±4.42),(44.45±3.91),(43.32±4.87)和(42.14±4.75)ng·L^-1;这6组的IL-6含量分别为(81.32±6.75),(124.22±8.31),(90.14±7.44),(119.23±8.42),(105.39±6.87)和(100.18±6.77)ng·L^-1。上述指标:模型组与假手术组相比,差异均有统计学意义(均P<0.01);对照组和高剂量实验组与模型组相比,差异均有统计学意义(P<0.05或P<0.01)。结论红芪总多糖可以通过改善脑缺血再灌注后脑组织神经元结构功能及通过调整促炎因子和抗炎因子的平衡,起到对脑缺血再灌注保护作用。
Objective To study the protection of total Hedysari polysaccharide on hippocampus of rats with cerebral ischemia-reperfusion injury.Methods Wistar rats were randomly divided into six groups:sham operation group,model group,control group,and experimental-L,experimental-M,and experimental-H groups,with15 rats in each group.The experimental groups were given total Hedysari polysaccharide in low,middle and high dose(50,100,200 mg·kg^-1).The control group was preoperatively administered 15 mg·kg^-1 nimodipine.The sham operation group and the model group were added with the same amount of 0.9%sodium chloride.The drug was administered once a day for 10 days.The model of cerebral ischemia-reperfusion injury was established by suture method in each group except sham operation group.The levels of serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10)were measured by enzyme linked immunosorbent assay.Results The levels of TNF-αin sham operation group,model group,control group,and experimental-L,experimental-M,and experimental-H groups were(35.46±4.31),(58.57±6.23),(39.88±5.46),(58.38±5.87),(56.43±6.27),(48.19±5.35)ng·L^-1,respectively;the levels of L-10 in the six groups were(35.24±4.22),(46.43±4.59),(38.72±4.42),(44.45±3.91),(43.32±4.87),(42.14±4.75)ng·L^-1,respectively;the levels of L-6 in the six groups were(81.32±6.75),(124.22±8.31),(90.14±7.44),(119.23±8.42),(105.39±6.87),(100.18±6.77)ng·L^-1,respectively.Comparison between model group and sham operation group,the difference of the factors were significantly(all P<0.01);comparison between control group,experimental-H group and model group,the difference of the factors were significantly(P<0.05 or P<0.01).Conclusion The total Hedysari polysaccharide can play a protective role in cerebral ischemia-reperfusion by improving the structure and function of neurons in brain tissue after cerebral ischemia-reperfusion and adjusting the balance of pro-inflammatory factors and anti-inflammatory factors.
