摘要:目的:通过改良差示酚硫法测定枸杞多糖含量,优化木瓜蛋白酶辅助提取枸杞多糖的工艺。方法:在改良差示酚硫法测定枸杞多糖含量基础上,采用均匀设计法,以枸杞多糖提取率为指标,木瓜蛋白酶用量、pH、酶解时间和酶解温度为考察因素,每个因素7个水平,建立4因素7水平的均匀设计试验,应用SPSS19. 0软件对数据进行多元回归分析,并通过相对黏度和体外抗氧化性对优化工艺下的枸杞提取物与传统提取比较。结果:改良差示酚硫法测定枸杞多糖条件为:测定波长λmax=484 nm,相关系数R^2=0. 9994,重复性RSD=2. 45%,回收率为98. 76%;木瓜蛋白酶辅助提取枸杞多糖的最佳工艺条件为:酶用量为0. 2%,pH为4. 2,温度为40℃,酶解时间为1. 0小时。木瓜蛋白酶辅助提取后,相同条件下枸杞提取物相对黏度,体外抗氧化性高于传统提取方法。结论:改良差示酚硫法可用于枸杞多糖含量测定,木瓜蛋白酶辅助提取枸杞多糖可提高其中多糖的利用率。
Objective: To optimize papain assisted extraction techonology for lycium barbarum polysaccharide(LBP) through determining the contents of LBP by improved differential phenol sulfuric method. Methods: On thefoundation of detecting the contents of LBP by improved differential phenol sulfuric method, four-factor seven-leveluniform design test was established by using the extraction rate of LBP as the index, the usage of papin, pH,enzymatic hydrolysis time and enzymatic hydrolysis temperature as the investigation factors, each factor in sevenlevels in uniform design method, multiple regression analysis of the data was made by SPSS 19.0 software, LBPextracted by the optimized technology and traditional extraction were compared. Results: The determinationconditions of LBP by improved differential phenol sulfuric method were: detection wavelength 姿max=484 nm,correlation coefficientR2=0.999 4, repeatability RSD=2.45%, recovery 98.76%; the optimal technology conditions ofextracting LBP by papain assisted extraction technology was: the dosage of enzyme 0.2%, pH 4.2, temperature 40℃,enzymatic hydrolysis time 1.0hour. After extracting by papain assisted extraction technology, under the same condi-tions, the relative viscosity and In-vitro antioxidant activity of LBP were higher than these of LBP obtained by tradi-tional extraction. Conclusion: Improved differential phenol sulfuric method could be used for content determinationof LBP, and papain assisted extraction techonology for LBP could improve the utilization rate of polysaccharide.
