摘要:目的通过中药慈菇消脂方干预高脂饮食诱导非酒精性脂肪性肝病(NAFLD)大鼠肝组织神经酰胺(Ceramide)Cer2和诱生型一氧化氮合酶(iNOS)信号通路的调节作用,探讨以解毒化痰法中药组方制剂的慈菇消脂方治疗NAFLD的作用机制。方法选用SPF雄性Wistar健康大鼠60只,随机分为造模组(50只)和正常对照组(10只),造模组通过高脂饮食成功建立NAFLD模型大鼠50只,随机分为模型组、慈菇消脂方高剂量组、慈菇消脂方中剂量组、慈菇消脂方低剂量组、吡格列酮西药对照组,每组10只。连续灌胃给药6周结束后,取材NAFLD大鼠肝脏和血清。检测肝组织、血浆中神经酰胺C2、肝组织游离脂肪酸(FFA)和iNOS,电镜下观察细胞器形态与结构变化。结果与正常组比较,模型组大鼠肝组织和血浆中神经酰胺C2、肝组织iNOS和FFA水平升高(P﹤0.01);电镜下肝组织呈中-重度脂肪变性并伴有肝细胞凋亡。与模型组比较,慈菇消脂方治疗组、吡格列酮对照组大鼠肝组织和血清神经酰胺C2,肝组织iNOS、FFA水平降低,(P﹤0.01)差异有统计学意义。结论以解毒化痰法组方的慈菇消脂方能够抑制NAFLD模型大鼠神经酰胺C2、iNOS、FFA水平,减轻肝细胞脂凋亡起到防治NAFLD的作用。
Objective To explore the regulatory role of ceramide C2 and iNOS signaling pathway in the liver of NAFLD rats induced by high-fat diet by Cigu Xiaozhi recipe, and to explore the effect mechanism of Cigu Xiaozhi recipe in the treatment of NAFLD. Methods Using SPF 60 healthy male Wistar rats, and randomly divided into modules(50) and normal control group(10), and successfully established by high fat diet 50, NAFLD model rats were randomly divided into model group, high dose Cigu Xiaozhi recipe group, medium dose Cigu Xiaozhi recipe group, low dose Cigu Xiaozhi recipe group, pioglitazone control group, each group of 10. Liver and serum samples were taken from NAFLD rats after 6 weeks of continuous gavage. The morphological and structural changes of organelles were observed by electron microscopy in liver tissues, plasma ceramide C2, liver FFA and iNOS. Results Compared with the normal group, the levels of ceramide C2, iNOS and FFA in liver tissues and plasma of model group were higher(P< 0.01).Under electron microscope, liver tissue showed moderate to severe steatosis accompanied by hepatocyte apoptosis. Compared with the model group, the liver tissue iNOS and FFA levels of rats in Cigu Xiaozhi recipe treatment group and pioglitazone control group decreased(P< 0.01).Conclusion The results showed that Cigu Xiaozhi recipe formulated with the method of detoxifying and resolving phlegm could inhibit the levels of C2, iNOS and FFA in rats with NAFLD,and inhibition of hepatic lipid apoptosis plays a role in the prevention and treatment of NAFLD.
