摘要:目的 :探讨去卵巢后大鼠骨髓上清中胰岛素样生长因子 1(IGF 1)和白细胞介素 1β(IL 1β)的变化及中药增骨丸对其影响。方法 :将 4 8只 8个月龄Wistar雌性大鼠随机分为正常对照组、模型组、乙烯雌酚组及 3种不同浓度的增骨丸组。每组 8只。正常组仅摘除一段肠系膜 ,其余 5组均做双侧卵巢切除术。术后 1周开始给药 ,连续用药 90d后取右侧股骨 ,剔净软组织 ,截取 1 5cm ,用 1mlPBS液将骨髓冲入DEPC处理过的试管中备用。分别测定骨髓中IGF 1和IL 1β的含量。 结果 :卵巢切除后大鼠骨髓上清中IGF 1的含量明显降低 ,而IL 1β的含量明显增高。经过雌激素及中药治疗能明显增高卵巢切除所致的IGF 1的降低 ,抑制IL 1β的增高 ,且其作用与剂量有关系。 结论 :增骨丸通过增加IGF 1的表达和降低IL 1β的表达而发挥防治骨质疏松症的作用。
Objective:To inquire into effects of Zenggu pill (ZGP) on bone medullae insulin-like growth factor-1 and Interleukin-1β in ovariectomized rats.Methods:Forty-eight healthy female Wistar rats were randomly divided into six groups:normal control group,model group,positive group and three ZGP groups (with high,moderate and low dosages respectively).The model group was excised a small part of mesentery and the other groups were ovariectomized bilaterally.They were put to death at the same time after three months.Then took right femur,measured the content of insulin-like growth factor-1 and Interleukin-1β in bone medullae by RIA and ELISA.Results:The content of insulin-like growth factor-1 in bone medullae of the high,moderate dosage of ZGP and positive control group rats were more higher than those in the model group (P<0.05),but the content of Interleukin-1β were much lower than that in the model group.Conclusion:The high and moderate ZGP can increase the content of insulin-like growth factor-1 and decrease Interleukin-1β in bone medullae.So they have the effect of prevention and treatment to postmenopausal osteoporosis,and the effects might be related to the level of doses of ZGP.
